Quality and in vitro fertilising ability of cryostored Pinzgau bull insemination dosesOriginal Paper

Applicability of cryopreserved insemination doses is dependent on the quality of the semen after thawing. The aim of the study was to check the options of new markers for the assessment of sperm quality to predict their fertilisation ability. In five bulls of the Pinzgau breed, the sperm fertilising ability was determined using the in vitro fertilisation. Bulls were divided according to the achieved blastocyst rate: bulls with good fertilising ability (NOB – 38.6%, KAZ – 28.9%, GAL – 29.3%) and bulls with low fertilising ability (LOH – 19.4%, NUS – 22.1%). In addition to the sperm motility and morphology, we monitored common physiological sperm characteristics: viability, apoptosis, acrosomal status, capacitation, mitochondrial activity and generation of reactive oxygen species using the flow cytometry procedure. Novel fertility-related biomarkers, such as ubiquitination, overexpression of MKRN1, SPTRX-3 and PAWP proteins or histone modification (H3K4me2), were also analysed by flow cytometry. From all monitored parameters, more proper characteristics of impaired in vitro fertilising ability proved to be high incidence of apoptotic markers (YO-PRO, Caspase 3/7) and higher counts of morphologically abnormal spermatozoa. Although the in vitro fertilisation (IVF) test can be an advantageous method for evaluating the sperm fertilising ability, there are still differences between the in vivo and in vitro fertilisation processes, which must be considered for example, when evaluating the sperm capacitation status.

Influence of daily feed ration on growth and condition of juvenile pikeperch (Sander lucioperca) reared in a recirculating aquaculture system (RAS)Original Paper

The study tested the effect of daily feed ration (DFR: 0.5%, 0.75%, 1.0%, 1.25%, and 1.5% of fish biomass) on juvenile pikeperch (Sander lucioperca) with an initial body weight of 21.5 ± 5.1 g and total length of 144.5 ± 8.5 mm. The pikeperch were fed floating feed at 8-hour intervals for a duration of 84 days. By the end of the experiment, the group fed DFR of 1.5% exhibited the highest body weight (51.5 ± 16.1 g) and total length (188.2 ± 17.8 mm). The weight heterogeneity, measured as the coefficient of variation (CV), ranged between 30.29 and 33.24%. The specific heterogeneity rate (SHR) ranged from 304.44 to 334.94‰/day. The group with DFR of 1.0% exhibited the highest degree of heterogeneity. Minor fin erosion was observed in the caudal fin by the end of the experiment. No significant differences were revealed in selected biochemical parameters indicating the liver, spleen, and intestinal function. All the fish tested were adequately fed, being provided sufficient nutrients for the proper growth of pikeperch. The DFR of 1.5% was evaluated as the most favourable. This amount of feed supported a higher number of values for Fulton’s coefficient (FC), specific growth rate (SGR), thermal growth rate (TGR), fish weight heterogeneity, growth of total fish biomass (BG), and optimum level of biochemical parameters in blood plasma.

Seaweed has an important role in the mitigation of enteric methane (CH4) production by ruminant animals. The utilisation and its effectiveness in enteric CH4 reduction require a preservation process. The aim of the study was to investigate the effect of different drying processes on the effectiveness of seaweed Gracilaria spp. in reducing CH4 production assessed through an in vitro method. Three drying techniques, sun-drying, oven-drying, and freeze-drying, were applied to produce a dried product of Glacilaria spp. Rice straw basal diets combined with concentrate at the 70 : 30% were used to test the inclusion of 4% of three differently dried products of Gracilaria spp. compared to the basal diet without seaweed (control group). Measurements were conducted on in vitro total and CH4 gas production, nutrient degradability, ammonia (NH3) and VFA concentration, and microbial population. Results showed that the three dried products of Gracilaria spp. significantly reduced in vitro CH4 production compared to the control group (P  0.05). All drying techniques gave a similar effect on in vitro CH4 reduction, but they did not affect dry matter (DM) and organic matter (OM) degradability (P  0.05) and significantly reduced neutral detergent fiber (NDF) degradability (P 0.05) compared to the control group, with oven-dried treatments having the lowest NDF degradability among the treatments. It is concluded that the three different drying techniques had similar effects on enteric CH4 reduction.

This experiment aimed to evaluate the effects of chitosan administration to total mixed ration (TMR) silage on ruminal fermentation characteristics and degradability by using both in vitro and in sacco methods. The completely randomized design (CRD) used in this study included five treatments and six replications. The treatments consisted of TMR silage with distilled water as control (SA), TMR with 1% acetic acid (SB), 0.5% chitosan in 1% acetic acid diluent (SC), 1% chitosan in 1% acetic acid diluent (SD), and 1.5% chitosan in 1% acetic acid diluent (SE). The variables observed were ruminal fermentation parameters, methane production and in sacco nutrient degradability. Results revealed that adding 1–1.5% chitosan (SD and SE treatments) reduced (P < 0.05) total protozoa, total gas production, methane production, the percentage of acetic acid, and the ratio of acetic acid to propionic acid, while it increased (P < 0.05) the proportion of propionic acid. The addition of chitosan (SE treatment) decreased (P < 0.05) the degradation of dry matter, but it increased (P < 0.05) the degradability of organic matter and crude protein, as determined by in sacco analysis.

Effects of maize silage withdrawal from finishing ration on weight gain, carcass yield and beef quality of Holstein Friesian × Belgian Blue crossbred bullsOriginal Paper

This study was practical work in a commercial beef cattle enterprise to offer beef producers different options in a total mixed ration (TMR) design. This study was conducted to determine the effects of withdrawing maize silage from TMR during the finishing period on weight gain, carcass yield and beef quality of meat in beef cattle. Fifty-two Holstein Friesian x Belgian Blue crossbred bulls were used in this study for 126 days. These bulls were divided into four feeding methods described as (i) no maize silage in TMR (C1), (ii) maize silage was withdrawn from TMR two months before slaughter (S1), (iii) maize silage was withdrawn from TMR one month before slaughter (S2), and (iv) maize silage was included in TMR until slaughter (C2). Results showed that the fattening performance of experimental bulls was not affected by treatments, except some colour parameters of the muscle. Also, its chemical composition, pH, water holding capacity, drip loss, thawing loss, cooking loss, shear force, thiobarbituric acid reactive substances and radical scavenging activity were not affected by treatments. To conclude, the withdrawal of maize silage from TMR did not affect fattening performance and meat quality, suggesting that there is no need to withdraw maize silage from TMR during the finishing period of fattening in beef cattle.

Abstract: Management of inbreeding is one of the crucial parts of breeding programs in livestock populations. Traditionally, the inbreeding coefficient is calculated using pedigree data; however, it can also be estimated from genomic data. Nowadays, various approaches to estimating genomic-based inbreeding coefficients are increasingly integrated into research and breeding practices. These genomic estimators can supplement or replace pedigree-based coefficients. Each genomic-based inbreeding coefficient has its own properties and different ranges of values, and some of them need specific settings for calculation. Moreover, depending on the methodological approach, genomic estimators are sensitive to the population structure, genotyping technology applied, and the quality control of obtained genomic data. It is important to consider all these factors when calculating and especially when interpreting the final genomic inbreeding values. For these reasons, using genomic-based inbreeding coefficients can be more challenging than using pedigree-based ones. In this review, we comprehensively evaluate the most commonly used genomic estimators of individual inbreeding in livestock, providing an in-depth analysis of their advantages and limitations while offering insights into the methodological considerations and best practices for their accurate calculation and interpretation.

Effect of tannin degradation of mangrove (Sonneratia alba) fruit on nutrient digestibility, protozoa population and methane gas productionOriginal Paper

This study examined the fermentation duration in relation to the effectiveness of tannin levels in mangrove (Sonneratia alba) fruit during the fermentation process using Aspergillus niger. The tested durations were 7, 10, 13, and 16 days in anaerobic conditions. The outcomes measured included crude fat, crude fibre, and nitrogen-free extract digestibility. Microbial biomass, protozoa population, and methane gas production were also recorded as responses to the rumen microbial activity. The results showed that 16 days of fermentation gave the highest average for all responses. The protozoa population reached around 4.07 × 105 cells/ml and methane gas amounted to 33.9 ml/g of dry matter (DM). This is caused by a decrease in the anti-nutrient tannin content in mangrove (S. alba) fruit due to fermentation by A. niger according to treatment. The conclusion of this research is that the 16-day fermentation treatment (T4) of mangrove fruit is the optimal time to be used as a source of concentrate feed for livestock in terms of the increase in nutritional value and gastrointestinal microbes represented by total protozoa population, crude fibre digestibility, crude fat digestibility, and the highest nitrogen-free extract digestibility. However, microbial biomass and methane gas production were not significantly different in this study.

Sheep milk production is a complex process that is influenced by various factors. This study aims to investigate how the litter size (single vs. twins), birth type (male vs. female), age of the ewes and body weight affect the milk composition and fatty acid profile of milk fat. Milk samples were collected from 119 Najdi ewes that were the subject of this study. Gas chromatography-mass spectrometry (GC-MASS) was used to quantify the fatty acids (FAs). The results showed that the twins birth type (female/female) has a significant influence (P < 0.05) on the concentration of linoleic acid (LA), alpha-linolenic acid (ALA), behenic acid (C22:0) and polyunsaturated fatty acid (PUFA). On the other hand, saturated fatty acids (SFA), docosahexaenoic acid (C22:4; DHA), and odd-chain fatty acids (OCFA), such as C15:0-antiso and C19:1-cis 10, increased significantly (P < 0.05) with the increasing age and body weight of the ewes, while the ALA and unsaturated fatty acids (UFA) significantly decreased (P < 0.05). The principal component analysis (PCA) revealed a positive association between the age and the OCFA, ALA and small-chain fatty acids (C6:0 and C8:0). In addition, the type of birth showed a positive association with the fat, lactose and palmitoleic acid C16:1 cis9. Conversely, there is a negative association between the UFA, monounsaturated fatty acids (MUFA) and PUFA. In addition, the body weight (BW) and litter size were negatively associated with the protein, SFA and medium-chain fatty acids (C10:0, C12:0, C14:0 and C16:0). The physiological factors generally suggested that the milk quality and essential FA, such as ALA, were influenced by the type of the lamb’s birth and the age of the ewes.

The fatty acid (FA) composition of milk fat can be positively influenced by the modification of dairy farming and the possible selection of raw material for processing. The question arises as to whether the benefits of a better FA composition will be maintained even after processing dairy products. Thus, the aim was to obtain a more favourable FA composition by a targeted selection of cow’s milk (grazing vs stabled herds) and then compare FAs of milk and fermented product (yoghurt). Bulk tank milk of grazing herds had significantly better FA composition than milk of stabled herds (for example, C16:0 28.07% vs 32.27%, P < 0.001; C18:3n-3 0.83% vs 0.41%, P < 0.001; C18:2 cis-9, trans-11 (conjugated linoleic acid; CLA) 1.02% vs 0.41%, P < 0.01). The differences between the FAs in milk and yoghurt samples were negligible (in relative values from 0.04% to 5.21%). The correlations between milk and yoghurt for nutritionally important FAs were high, from 0.925 0 (C18:2n-6) to 0.998 8 (CLA; both P < 0.001). The minimal effect of milk fermentation on the original FA composition of milk fat was found. In conclusion, systematic selection of raw cow’s milk or modification of farming conditions can also provide a nutritionally desirable composition of final dairy products.

Cryopreservation of early-stage male Black Sea trout (Salmo trutta labrax) germ cells and comparison of the whole tissue and enzymatically isolated cellsOriginal Paper

The high plasticity, or the ability to differentiate into various cell types, and capacity to become both gametes of early-stage germ cells (eGCs) allows them to be used for the long-term storage and recovery of genetic information. In this study, species-specific protocols (isolation period, enzymatic separation solution, incubation period, appropriate cryoprotectant, and cryopreservation protocol) were established for the isolation and long-term preservation of Black Sea trout eGCs. In addition, the difference between the application of cryopreservation to the whole gonad tissue and to the enzymatically separated cells has been shown in terms of viability and the number of cells obtained. According to the cell count made from the histological sections of the testicular tissue throughout the year, the period with the highest concentration of eGCs in the tissue is between May and June. To identify the optimal enzymatic dissociation solution, we subjected testicular tissues to digestion using various combinations of trypsin (T) and collagenase (C) (0.3–0.5% T and 0.1–0.3% C). Our findings revealed that the 0.3% C provided the highest yield of viable cells (90.9%). We cryopreserved the enzymatically dissociated cells (EDCs) using six different cryoprotectants (Dimethyl sulfoxide, Glycerol, and CryoSOfree™, in both L-15 and phosphate-buffered saline – PBS mediums). Additionally, whole tissue exclusively with CryoSOfree™. After storing the samples at –152 °C for two years, the group of EDCs cryopreserved in L15+Glycerol exhibited the highest viability at 93.3%. Meanwhile, the whole tissue group yielded the greatest number of cells per ml after thawing, with a count of 33 210 000 cells. When considering both the viability (91.11%) and the number of cells obtained (33 210 000 cells/ml), the whole tissue cryopreservation group outperformed all the other methods.

Negative energy balance (NEB) after calving leads to unique metabolic changes in cows and it provides important information on the period when nutrient output prevails over nutrient intake associated with a number of physiological changes in the organism suitable for detection using biochemical blood analyses. The aim of this paper is to summarise current knowledge of the changes in blood parameters during the period of NEB in relation to non-esterified fatty acids, ketone bodies, and immunologic and stress indicators. The impact of these changes on bovine reproduction and health is also discussed in the sequel. The period of NEB is still a very pertinent area of research as it provides a new insight into connections between physiological systems, to wit, ​​immunology, manifestation of various stress indicators, including oxidative stress, and heat shock proteins inter alia. Understanding the physiological changes during NEB is crucial for successful management of the transition period and subsequent overall good animal health and productivity.

The effect of different forages on rumen microbiota and milk production performance in Holstein dairy cowsOriginal Paper

Optimising the feed composition, especially the forage choice, and reducing costs are essential for improving dairy production efficiency. Different forage sources and proportions were used to formulate rations containing equal energy and protein, and their effects on rumen microbiota and milk production performance of dairy cows were evaluated in two experiments. In experiment 1, thirty lactating cows (235 ±13 d postpartum; milk production 29.1 ± 1.0 kg/day) were divided into Pangola and Bermuda groups. In experiment 2, twenty lactating cows (79.9 ± 8.1 d postpartum; milk production 34.7 ± 0.6 kg/day) were divided into Pangola and oat groups. In experiment 1, the Simpson index for rumen microbiota of the Pangola group was significantly higher than in the Bermuda group (P < 0.05). Analysis of the weighted unique fraction (UniFrac) distances indicated significant differences in the beta diversity of the community composition of rumen microbiota between Pangola, Bermuda and oat groups in both experiments (P < 0.001). The relative abundance of Prevotella brevis was significantly higher in the Pangola group than in the oat group in experiment 2 (P < 0.05). The somatic cell counts (SCCs), C18:0, and C18:1 in milk were significantly higher in the Bermuda group than in the Pangola group (P < 0.05) in experiment 1. On the other hand, milk crude protein (CP) and solids-not-fat (SNF) were significantly higher in the Oat group than in the Pangola group (P < 0.05) in experiment 2; however, milk urea nitrogen (MUN) was significantly higher in the Pangola group (P < 0.05). In conclusion, a switch of forage (Pangola vs Bermuda) at a lower proportion of the diet under the high forage level condition (experiment 1) caused only minor changes in rumen microbiota diversity (Simpson index, beta diversity) and milk production performance (milk SCCs, C18:0 and C18:1). On the other hand, a switch of forage (Pangola vs oat) at a higher proportion of the diet under the low forage level condition (experiment 2) resulted in greater changes in rumen microbiota diversity (beta diversity, relative abundances of bacterial taxa, P. brevis relative abundance) and milk production performance (milk CP, SNF, and MUN).

Livestock farming is vital to a country's economy, and technological innovations in animal genetics and reproductive biotechnologies are key for environmental and socioeconomic development. This study aimed to evaluate the antioxidant capacity of Coffea arabica grounds (CAG) extract on the semen parameters of Fleckvieh cattle. CAG was processed, and its antioxidant capacity was assessed using DPPH, FRAP, and total phenols assays. Semen was collected from a pedigree bull and analysed macroscopically and microscopically. Different concentrations of CAG (0, 1, 1.5, and 2 mg/ml) were tested at 4, 12, 24, and 36 h post-refrigeration. The study used a completely randomised design with ten replications, and variables such as motility, kinetic parameters, membrane functionality, and acrosomal integrity were analysed. The normal distribution of the variables – motility, kinetic parameters, membrane functionality, and acrosomal integrity – was analysed using the Shapiro-Wilk statistical test (P > 0.05). An analysis of variance was then performed with a significance level of P < 0.05 to compare the means, using InfoStat software. Results showed that 1 mg of CAG significantly improved total motility and progressive motility at 4 h, along with kinetic parameters and membrane integrity. Overall, CAG exhibits antioxidant properties that enhance sperm characteristics, particularly at 1 mg/ml concentration after 4 h of refrigeration. However, further studies are needed to understand better the mechanisms of action and the overall efficacy of CAG as an antioxidant agent in this specific context.

Does the incidence of egg yolk influence the meat quality and fatty acid profile of broilers of two chicken genotypes?Original Paper

The present experiment studied the effects of egg composition (egg yolk content; Y) of two different chicken genotypes (selected by computed tomography; CT) on hatched chick growth performance, slaughter traits, and meat quality. Three thousand five hundred eggs per genotype were scanned by CT. Then, for each genotype, eggs were selected according to their Y content: low (21.0 ± 0.88 – 350 eggs), medium (24.5 ± 0.15– 350 eggs) and high (28.3 ± 0.98 – 350 eggs). The remaining eggs were excluded from the study. At 11 weeks of age, 15 chickens per group were slaughtered, carcasses were dissected and breast and legs were excised and dedicated to meat quality evaluations. Many parameters were influenced in the meat-type (EE) chickens, including slaughter, breast and leg weights, and abdominal fat content, whereas only the breast incidence on slaughter weight was affected in Tetra-H hybrid. In neither genotype were the meat traits affected by Y content. Two exceptions were represented by leg thawing loss, higher in low Y group compared to medium and high Y group, and by tibia length, longer in high Y content group than in the medium one, both for EE chickens. Similar findings were observed for the meat fatty acid (FA) profile, as only some single FA were influenced by Y content, however, apparently without a specific physiological meaning. The present work demonstrated that the Y content, measured by CT, allowed to select high-quality meat-type animals characterised by the best productive performances in terms of growth rate, live weight, slaughter weight, breast and thigh weights, and with lower abdominal fat content. These findings would bring positive advantages to hatcheries in terms of chick quality and also to farmers in terms of economic revenues: They would rear robust animals that would guarantee a higher probability of survival in the first rearing period and would be characterised by a high slaughter weight at the end of the productive cycle.

The development of intelligent systems for the tracking of free-range livestock is a challenge to both information and communication technology (ICT) scientists and those in the animal sciences. Cyber-physical systems make it possible to track and control processes involving intelligent objects from the physical and virtual worlds. In the case of free-range grazing, it is necessary to manage processes in two domains ‒ that of the intelligent pasture management and that of the animals. Due to the differences in the conditions of different types of pastures – plain or high land and the characteristics of the cattle breeds, ready-made models cannot be used, but it is necessary to build a specific multi-aspect model for the behaviour and life cycle of cows. Our team organised their research on cows from two different breeds (Rhodope Shorthorn Cattle and Bulgarian Rhodope Cattle) raised in similar technologies, grazed on two different types of pasture. The aim of the study is to develop a comprehensive model for determining cattle behavioural activities on pastures using sensor groups, by incorporating physical observations and appropriate statistical models.

This study compared the efficacy of the egg yolk (EY) and soy lecithin (SL) semen extender during 72-h storage at 4 °C in the presence/absence of streptomycin, penicillin, lincomycin, and spectinomycin. Bull semen samples (n = 12) were obtained and stored in either ANDROMED® (SL extender) or TRILADYL CSS® (EY extender). After 24, 48, and 72 h of storage at 4 °C, bacterial colonies (CFU – colony forming units) were grown and counted on tryptic soy and blood agar, and the bacterial isolates were identified using the MALDI-TOF MS Biotyper. Sperm motility, mitochondrial membrane potential (MMP), membrane and acrosome integrity, sperm DNA fragmentation, and reactive oxygen species (ROS) production were analysed. The results showed that the type of semen diluent could be crucial for the efficacy of used antibiotics, since significantly decreased bacterial occurrence following 48 h or no bacterial growth after 72 h was recorded in the SL-based groups. Even the SL medium itself did not favour bacterial growth, as significantly decreased bacterial load was observed after 48 h (< 0.01) and 72 h (<0.000 1). In contrast, the bacterial load in the EY medium without antibiotics significantly increased (< 0.05). In contrast, an improved ability to preserve sperm quality parameters was observed in EY-extended semen. In particular, sperm motility was significantly higher (< 0.000 1 and < 0.001) after 24 h in each EY-based group in comparison to the control (Ctrl), whereas among the SL-based groups, only the spectinomycin-supplemented group presented with a significant motility improvement (< 0.01) when compared to the Ctrl. Although the EY semen extender provided enhanced sperm quality preservation during the 72-h storage, in the future, the SL medium composition should be improved to match the sperm preservation ability of the EY medium while maintaining its microbial safety.

This study was conducted to investigate the effects and mechanisms of glutamine (Gln) on the repair of acid-induced injury in dairy cow ruminal epithelial cells (RECs) in vitro. Dairy cow RECs were cultured in a medium with pH of 5.5 for 3 h and subsequently treated with various concentrations of Gln (4, 8, 12, 32 mmol/l) for 12 h. The messenger ribonucleic acid (mRNA) expression levels of occludin (OCLN), claudin 1 (CLDN1), toll-like receptor 2 (TLR2), toll-like receptor 4 (TLR4) and genes for inflammatory factors were measured using reverse transcription-quantitative real-time PCR (RT-qPCR). The results showed that cellular activity and OCLN expression were significantly highest at 8 mmol/l Gln (< 0.05). CLDN1 expression was significantly higher at 4 mmol/l Gln compared to the other groups (< 0.05). The relative expression levels of tumour necrosis factor (TNF), interleukin 1B (IL1B), C-X-C motif chemokine ligand 8 (CXCL8), TLR2 and TLR4 in the acid treatment group were significantly higher than those in the control group (< 0.05), but they were lower in the Gln-treated groups than in the acid treatment group (< 0.05). These findings demonstrate that Gln promotes the proliferation of RECs, enhances the expression of epithelial cell junction proteins, and inhibits the expression of inflammatory factors and surface receptors. In conclusion, Gln shows a potential for repairing acid-induced injury in RECs.

Dietary antioxidant supplementation has become popular for managing male infertility because of its impact on the antioxidant capacity of semen. This study evaluates the effect of dietary antioxidant supplementation on the seminal characteristics of Colombian Creole horses. Semen from six Colombian Creole stallions was collected using an artificial vagina. The stallions were randomly allocated to a control group (CG) or a supplementation group (SG) that received a multi-antioxidant supplement. A repeated measures design, with a wash out period of two weeks between treatments, was used. Sperm motility and kinematics were assessed using a computer-assisted semen analysis system, membrane integrity (MI) and the acrosomal integrity (AI) of sperm were assessed using fluorescent microscopy, abnormal morphology (AM) was analysed using supravital staining, and the functionality of the sperm membrane was assessed using the hypoosmotic swelling (HOS) test. A fraction of each ejaculate sample was centrifuged to obtain seminal plasma (SP). The reactive oxygen species and the total antioxidant capacity (TAC) of the SP were measured via spectrofluorimetry. Linear models were fitted, and comparison of means was performed using Tukey’s test. Dietary antioxidant supplementation increased sperm beat cross frequency. In addition, a higher AI was found for the SG. Supplementation also increased the TAC of the SP. Individual analysis revealed that dietary antioxidant supplementation increased the total and progressive motility of the semen of two horses in the SG; however, supplementation reduced these two parameters in two other individuals compared to the CG. It can be concluded that dietary antioxidant supplementation with a nutraceutical that provides vitamins, minerals, amino acids, and omega-3 for horses can increase sperm quality as well as the TAC of SP; however, this may be subject to the individual effect of each horse.

This research evaluated the suitability of commercially available larval feeds, Otohime B2 (OB2), Aller Infa (AI), and Aqua Start (AS), and one Experimental Feed (EF), for the weaning of largemouth bass (Micropterus salmoides), LMB. Feeds were presented with various ω-3 fatty acid levels/bioavailability (high in OB2 and AI), fat percentage (high in OB2 and AS), free amino acid and short peptide (FAA + SP) levels (high in OB2), and various soluble protein (SPR) levels (high in AS and EF). Fish were co-fed Artemia plus OB2 from the 19th to 22nd day post-hatching (DPH), then Artemia in addition to one of the four above diets for seven days, with complete Artemia removal on the 30th DPH. Fish were sampled on the 32nd DPH. Morphometry, digestive enzyme activities, hormonal status, skeleton, muscle development, and potentially pathogenic Flavobacterium spp. levels were estimated. Survival was high (96% or more) in all the weaning regimes. Weaning to OB2 was linked to a fast fish growth rate (14.29%/day), while both OB2 and AI supported the skeleton development. Weight gain correlated with total fat, ash levels, free amino acids, and short peptide levels in the diet. Larvae weaned to soluble protein-rich AS and EF showed the lowest fish weight gain and skeleton development, and lower growth of potentially pathogenic Flavobacterium spp. This research suggests that the weaning diets for largemouth bass should have a balanced protein content and quality while allowing for the inclusion of fewer marine ingredients.

Effect of dehulled lupin seeds in feed mixture on muscle protein quality of broiler chickensOriginal Paper

The aim of the study was to prepare and test the effect of diets with 50% and 100% replacement of soybean meal with dehulled seeds of white lupin (Lupinus albus) variety Zulika. Two hundred and forty ROSS 308 broiler chickens were divided into control group (C) and two experimental groups E50 (50% replacement of soybean meal with dehulled white lupin meal) and E100 (100% replacement of soybean meal with dehulled white lupin meal). The production parameters and the amino acid spectrum of breast and thigh muscles were evaluated up to 34 days of age. Final live weight of broiler chickens was not significantly affected by the substitution of soybean meal in starter, grower and finisher diets (C 2.3 ± 0.3 kg, E50 2.4 ± 0.3 kg, E100 2.4 ± 0.3 kg). Lupine meal-based diets positively influenced chicken health, which was reflected in a lower mortality rate in the experimental groups (E50 5.0 %, E100 2.5 %) compared to the control group (C 7.5 %). Differences in feed conversion were minimal (C 1.47 kg/kg, E50 1.48 kg/kg, E100 1.45 kg/kg). When evaluating the total amino acid content in the breast and thigh muscle of broiler chickens of the experimental groups (E50, E100), it can be noted that compared to the C group, the total contents of both essential and non-essential amino acids were significantly (P ≤ 0.05) lower in the dry matter of thigh muscle.

Industrial dairy farms boast highly advanced health monitoring and disease diagnosis systems. But without easily accessible, user-friendly web platforms for real-time decision-making, most dairy farmers cannot proactively manage herd health management and optimize treatments based on disease prediction and prevention. To bridge this gap, we have developed a web application of a Decision support system (DSS) for dairy health management based on machine learning. The system architecture combines a Flask backend with a React frontend and scalable cloud data storage and includes preprocessing, data integration, predictive modelling, and cost analysis. DSS forecasts herd diseases with an accuracy 6.66 mean absolute error and 2.35 median absolute deviation across predictions. Its core predictive capabilities rely on long short-term memory (LSTM) neural networks to forecast disease progression from historical records and on a linear trend model to project cuts in treatment costs. The system calculates medication dosages and cost per disease, streamlines supplier selection, and simulates various treatment scenarios, thereby identifying high-cost diseases with potential savings. In other words, this DSS application processes disease and treatment data by incorporating veterinary records into advanced data analytics and neural networks, thereby predicting diseases, optimizing disease prevention and treatment strategies, and reducing costs. As such, this DSS application provides dairy farmers with a tool for strategic decision-making, veterinary treatment planning, and cost-effective disease management towards improving animal welfare and increasing milk yield.

Mastitis, mainly caused by contagious bacteria, is an important disease in dairy goat production, especially subclinical mastitis. The aim of the research was to study the relationship between somatic cell count (SCC) and presence of mastitis pathogens in goat milk during the period 2022–2024. A total of 987 udder-half milk samples were obtained from 5 goat farms in Slovakia. The presence of pathogens was determined by bacterial cultivation and grown colonies were identified by MALDI-TOF MS at the species level. Fossomatic 7 was used to determine SCC. To evaluate the frequency distribution of the samples, the following groups of SCC were considered: SCC1 < 500 × 103 cells/ml, SCC2 ≥ 500 < 1 000 × 103 cells/ml, SCC3 ≥ 1 000 < 2 000 × 103 cells/ml, SCC4 ≥ 2 000 × 103 cells/ml. The individual milk sample were 42.2% in SCC1, 17.0% in SCC2, 16.7% and 24.0% in SCC3 and SCC4, respectively. Of the total number of samples, 23.0% were bacteriologically positive. The most frequently identified pathogens were Staphylococcus spp., especially non-aureus staphylococci and mammaliicocci (NASM). Among the NASM, Staphylococcus epidermidis (40.3%), Staphylococcus caprae (27.5%) and Staphylococcus simulans (10.4%) were the most prevalent species. Only 2 samples were positive for Staphylococcus aureus. A higher occurrence of pathogens was identified in SCC ≥ 1 000 × 103 cells/ml (SCC3 and SCC4) compared to SCC < 1 000 × 103 cells/ml (SCC1 and SCC2). However, approximately 34.8% of uninfected half udders with no bacteriological findings had SCC ≥ 1 000 × 103 cells/ml. In conclusion, NASM were the most common bacterial causative agents of subclinical mastitis in goats and it is not easy to set a specific SCC for subclinical mastitis diagnosis as it is in dairy cows.

Effect of dehulled lupin seeds in feed mixture on muscle protein quality of ducklingsOriginal Paper

The aim of the study was to prepare and test the effect of diets with 50% and 100% replacement of soybean meal with dehulled seeds of white lupin (Lupinus albus) variety Zulika. One hundred and eighty Cherry Valley duckling were divided to control group (C) and two experimental groups E50 (50% replacement of soybean meal with dehulled white lupin meal) and E100 (100% replacement of soybean meal with dehulled white lupin meal). There were evaluated the production parameters and the amino acid spectrum of breast and thigh muscles up to 42 days of age. The effect of diets containing lupin meal had no negative effect on the yield of duckling. In particular, positively can be evaluated higher growth rate in the experimental groups compared to the control (C 3.22 ± 0.28 kg, E50 3.40 ± 0.27 kg, E100 3.48 ± 0.28 kg), at the same time significantly lower mortality in the experimental groups compared to the control (C 5.88%, E50 1.47%, E100 1.49%) and, except for E100, minimal differences in feed conversion ratio (C 2.33 kg/kg, E50 2.27 kg/kg, E100 2.71 kg/kg). Feeding diets based on lupin meal resulted in the reduction of most essential and non-essential amino acids (P ≤ 0.05) in the dry matter content of both breast and thigh muscle (E50, E100). Lupine meal-based diets positively influenced ducks’ health, which was reflected by lower mortality rate in the experimental groups. Therefore, the results show that diets based on lupin meal had a positive effect on the health and welfare of intensively fattened ducks and that they affected positively the economic aspect of breeding.

Whole-genome resequencing data reveal the genetic diversity of local chickens in southern Zhejiang and surrounding areas in ChinaOriginal Paper

China’s local chicken breeds, especially those in the Zhejiang Province, are rich in genetic diversity owing to environmental heterogeneity and complex ecosystems. Distinctive local breeds have emerged through long-term natural selection and domestication. We investigated the genetic diversity and population structure of local chickens in southern Zhejiang and surrounding areas using whole-genome resequencing of 129 chickens from seven populations. A total of 1.8 terabytes of raw data was obtained, and 4 802 728 single nucleotide polymorphisms were detected. The Xianju chicken population exhibited the highest genetic diversity, while Yandang Partridge chickens were genetically distant from other chicken breeds. This study provides valuable information for conserving poultry genetic diversity and informs about breeding programmes of local Chinese chicken breeds.

Impact of microclimatic conditions on sperm production in Czech Holstein bulls: A correlational studyOriginal Paper

In this study, the influence of microclimatic conditions in different seasons of the year (including the hottest months) on the quantitative and qualitative characteristics of ejaculates of Czech Holstein bulls was assessed. Bulls were kept in the facility with no forced ventilation or air conditioning. To determine the influence, the temperature-humidity index (THI) was calculated based on the microclimatic parameters measured directly in the facility with the animals. Bull sperm was obtained using an artificial vagina on five occasions throughout the year, in different seasons of the year including the hottest months. Semen was assessed for its volume, sperm concentration, and motility by CASA, as well as cellular viability by using flow cytometry. In the present study no statistically significant positive correlations between values of THI and sperm degradation were observed. Obtained preliminary results give grounds to believe that Czech Holstein bulls produced high-quality sperm during the summer, the hottest months of the year.

Mesobiliverdin IXα-enriched microalgae feed improves gut health in weaned piglets under LPS challengeOriginal Paper

This study aimed to evaluate the effect of mesobiliverdin IXα-enriched microalgae (MBV-SP) on growth, diarrhoea prevalence, intestinal morphology, antioxidant capacity, and inflammatory cytokines in lipopolysaccharide (LPS)-induced enteritis in weaned piglets. Seventy-two 28-day-old piglets were randomly divided into four groups: control (NC), LPS, tylosin, and MBV-SP, with saline/LPS (100 µg/kg) administered intraperitoneally on day 14. Results showed reduced mortality and diarrhoea rates in the tylosin + LPS and MBV-SP + LPS groups compared to the LPS group. Histological analysis revealed improved villus morphology and decreased crypt depth in the MBV-SP group. Catalase activity was notably higher in the MBV-SP group. Both tylosin and MBV-SP supplementation reduced pro-inflammatory cytokines, with MBV-SP exhibiting a more pronounced effect. In conclusion, MBV-SP mitigated diarrhoea, improved antioxidant capacity, and modulated inflammatory cytokines in weaned piglets with LPS-induced enteritis, highlighting its potential as a dietary supplement for the gut health in piglets.

Effects of two protein levels on the performance of chicken males with different growth intensitiesOriginal Paper

At present, genetic selection programs produce chicken genotypes with different growth intensities, which might have variable requirements for dietary protein. The objective of this study was to compare the response of three different genotypes to two levels of crude protein in feed mixtures. Cockerels of fast-growing Ross 308, medium-growing Hubbard JA 757 and slow-growing ISA Dual chickens were used in the study. Each genotype was fed diets that differed in protein level: the control group (C) received commercial feed, and the experimental group was fed a diet with a 6% lower protein content (LP). The daily weight gain (DWG) and feed conversion ratio (FCR) were significantly affected by the interaction of genotype and feed protein level. A greater percentage of DWG depression was observed in fast-growing cockerels than in medium-growing cockerels (10% and 6%, respectively), whereas the percentage of slow-growing cockerels negligibly increased (2%). A low-protein diet impaired the FCR only in fast-growing birds (–5%), whereas in the medium- (–2%) and slow-growing groups (+2%), the differences were not significant. Carcass composition significantly influenced only genotype and thigh meat pH. In terms of meat colour, significant interactions revealed that in fast-growing Ross 308 chickens, redness and yellowness did not differ according to diet group; however, in both genotypes with slower growth, significantly greater redness and yellowness were detected in the low-protein diet group than in the control group. The results indicate that genotypes with slower growth have lower protein requirements for growth performance, but lower diet protein has an effect on physical meat quality parameters in these genotypes.

Genomic diversity characteristics of sheep populations in the Czech Republic, Slovakia, Serbia and MontenegroOriginal Paper

Monitoring the genetic diversity in livestock is a critical component for sustainable management of small and endangered breeds. In this paper we follow up the two most often used metrics of genetic diversity, the average inbreeding coefficients (F) and the effective population (Ne) size in 16 sheep breeds from four countries. The analysis was based on single nucleotide polymorphism (SNP) markers, with about 35 to 49 thousand SNPs and 19 to 98 individuals after quality control. The runs of homozygosity (ROH) method with the cgaTOH software was used to estimate FROH, for three different time points in the past (3, 6 and 12 generations before present). Three methods were implemented to estimate Ne, using the NeEstimator v2, GONE and CurrentNe software for different time spans in the past. The average FROH ranged between 0.001 and 0.035 for 3 generations, 0.003 and 0.059 for 6 generations, 0.005 and 0.074 for 12 generations in the past. The wide range of inbreeding coefficients might be a consequence of different population sizes and breed management strategies in the respective countries. The results for Ne ranged from 7 to 352 for NeEstimator, 25 to 303 for GONE, and 15 to 542 for CurrentNe. Overall, the software showed comparable results for 10 out of 16 breeds, and significantly different results for at least one software for at least one of the methods for 6 out of 16 breeds. These differences show a degree of uncertainty in Ne estimations for certain breeds. Overall, we suggest the continued monitoring of sheep breeds, ideally by routine genotyping in all populations. This monitoring is especially important for small breeds with a greater decline in genetic diversity.

Estimation of lean meat percentage in pig carcass with the use of objective methods with regard to sexOriginal Paper

In the Czech Republic, the pig carcass classification is mandatory in slaughterhouses processing over 200 pigs weekly. As breeding practices evolve to enhance lean meat yield, it is essential to update regression equations used in classification systems. This study presents new regression models for the Fat-O-Meater II (FOM II) device, using computed tomography (CT) as the reference method. Separate equations were developed for barrows, gilts, and boars to improve the accuracy of lean meat percentage (LMC) estimation. To calibrate the CT method, 24 carcasses were selected across a range of backfat thicknesses and sexes. CT scans were performed on chilled left carcass halves, followed by manual dissection to determine the true LMC. A correction model was applied to align the CT-derived LMC with dissection results. Subsequently, 128 carcasses were measured using FOM II and CT to develop sex-specific regression equations using ordinary least squares. The models revealed sex-specific differences in prediction accuracy. Gilts achieved an R2 of 0.66 and RMSEP of 1.35; barrows had higher R2 (0.759) and greater RMSEP (1.46); boars showed the most consistent composition (R2 = 0.734, RMSEP = 1.14). Compared to the standard method, gilts and boars had slightly higher LMC (+0.03% and +0.82%), while barrows had lower LMC (–0.14%). These differences translated into economic impacts, with gains of CZK 1.23 and CZK 4.33 per gilt and boar carcass, respectively, and a loss of CZK 5.55 per barrow carcass. These results support the formulated hypotheses, and the fact that sex-specific calibration enhances classification accuracy and economic efficiency.

This study aimed to evaluate the effects of silymarin supplementation during late gestation on reproductive performance, haematological parameters, antioxidant capacity, and gut microbiota composition in sows. Twenty parity-4 crossbred sows (Landrace × Yorkshire) were enrolled and randomly allocated in parity blocks to either a control group (CG; n = 10, basal diet) or a silymarin-supplemented group (SIL; n = 10, basal diet + 200 mg/kg silymarin). The experimental period extended from day 85 of gestation to the completion of farrowing. The results demonstrated that dietary silymarin significantly reduced the number of stillbirths (P < 0.05), without exerting a significant effect on the total number and proportion of live-born piglets (P > 0.05). No notable differences were observed in haematological parameters between the two groups (P > 0.05). However, catalase (CAT) activity and total antioxidant capacity (T-AOC) were significantly elevated in the silymarin group (P < 0.05), and superoxide dismutase (SOD) showed a tendency to increase (P = 0.078). High-throughput 16S rRNA sequencing revealed 1 671 unique feature sequences in the silymarin group and 1 073 in the control group, with 1 600 sequences shared between the two groups. A trend towards increased dominance was observed in the silymarin group (P = 0.082), while both the Shannon and Simpson indices tended to decline (P = 0.087; P = 0.082), suggesting a possible reduction in microbial diversity. Principal coordinate analysis (PCoA) of β-diversity revealed significant structural differences in gut microbiota between the two groups. SIMPER analysis identified Terrisporobacter as the principal genus contributing to these differences. In conclusion, silymarin supplementation during late gestation may enhance reproductive outcomes in sows, potentially through modulation of gut microbial composition and enhancement of systemic antioxidant status.
