Anim. Sci., 2025, 70(8):319-334 | DOI: 10.17221/90/2025-CJAS The somatic cell count (SCC) in raw milk is an important indicator of health and hygienic quality. Artificial reduction of the SCC (ARSCC) in milk, for the apparent improvement of milk quality for commercial reasons, is an undesirable phenomenon and a violation of authenticity both in the factual sense and legislatively. Analytical methods need to be developed to identify ARSCC as well as to assess the effects that ARSCC may have on milk. The aim of the work was to quantify the effects of ARSCC on cow’s milk as a food raw material. The results presented are some of the first on the given problem. Raw bulk tank cow milk was sampled in two experiments, each time for the whole year (2021–2022 and 2023–2024, n = 66 and n = 53, respectively) from herds of Czech Fleckvieh and Holstein cows, 1 : 1. ARSCC in experiment 1 (n = 66) slightly reduced the fat content, in experiment 2 (n = 53) it did not, otherwise the milk indicators with the exception of SCC were almost (1) and completely (2) identical. All samples were negative for the presence of inhibitory substance residues. ARSCC under the specified technological conditions reduced SCC (1) from 772 ± 906 103/ml to 376 ± 630 103/ml, by –51.3% (P < 0.001) and (2) from 592 ± 798 103/ml to 304 ± 468 103/ml, by –48.5% (P < 0.001). Under these circumstances, the milk fermentation ability improved (1) from 28.52 ± 4.72 °SH to 31.0 ± 4.65 °SH, by 8.66% (P < 0.001) and (2) from 32.51 ± 2.61 °SH to 33.80 ± 2.88 °SH, by 3.97% (P < 0.001). Curd firmness was better for SCC ≤ 400 103/ml compared to higher SCC by 5.26% (P < 0.001). Nevertheless, it is not possible, for hygienic and health reasons, to allow such milk for human consumption and it is necessary to find effective identification analytical methods for ARSCC. |

However, the use of biofortified feedstuffs remains a not fully realized strategy to enhance the Se content of animal derived products. This study explored an on-farm biofortification approach by incorporating Se-enriched maize silage into the total mixed ration (TMR) of dairy cows. Sixty Holstein cows were divided into a control group (CON), receiving a conventional diet with selenite supplementation (0.6 mg/kg Se in TMR), and an experimental group (EXP), in which conventional silage was replaced with high-Se silage (0.9 mg/kg Se in TMR). The trial lasted 22 weeks, including one week of adaptation and four weeks after supplementation, when Se concentrations in milk, Se transfer efficiency, and key milk components critical for the production of Se-enriched dairy products were assessed. The higher Se concentration in the TMR had no adverse effects on milk composition or antioxidant status. Milk Se concentration in the EXP group increased rapidly, reaching 68 µg/l within two weeks, significantly higher (P < 0.005) than in the CON group (27 µg/l). Se transfer efficiency to milk was also higher in the EXP group (13.9%) compared to the CON group (8.8%). The diverse Se species in biofortified silage, confirmed through the speciation analysis, may have contributed to these outcomes. However, the gradual decline in milk Se after the initial peak warrants further investigation into physiological factors or changes in silage Se speciation during storage. |

Effect of cattle breed on muscle protein qualityOriginal PaperEva Straková, Natalie KlessyCzech J. Anim. Sci., 2026, 71(5):220-226 | DOI: 10.17221/55/2026-CJAS The aim of the study was to compare the muscle protein quality in the m. longissimus, pars thoracis of bulls of the Aberdeen Angus (AA) and Czech Fleckvieh (CF) breeds reared under identical conditions within an extensive pasture-based system. The CF breed exhibited a higher crude protein content (887 ± 87.4 g/kg of dry matter) compared to AA (812 ± 59.3 g/kg of dry matter); P ≤ 0.05. A higher content of non-essential amino acids was recorded in CF (390 ± 37.9 g/kg of dry matter) compared to AA (364 ± 44.5 g/kg of dry matter); P ≤ 0.05, as well as higher contents of essential amino acids (CF 406 ± 38.9 g/kg of dry matter, AA 382 ± 41.0 g/kg of dry matter; P ≤ 0.05) and total analysed amino acids (CF 796 ± 74.8 g/kg of dry matter, AA 746 ± 82.7 g/kg of dry matter; P ≤ 0.05). Breed-related differences were observed in the content of most analysed amino acids (P ≤ 0.05), except for tyrosine and phenylalanine, for which no effect of breed was found (P > 0.05). Lysine showed the highest content among essential amino acids (AA 73.4 ± 8.23 g/kg of dry matter, CF 76.6 ± 8.87 g/kg of dry matter; P ≤ 0.05), while glutamic acid was dominant among non-essential amino acids (AA 109 ± 14.4 g/kg of dry matter, CF 119 ± 12.5 g/kg of dry matter; P ≤ 0.05). The ratio of essential to non-essential amino acids (EAA/NEAA) did not differ between breeds (AA 1.05 ± 0.08, CF 1.04 ± 0.05; P > 0.05). The results indicate that cattle breed affects the content of crude protein and the content of most analysed amino acids in the muscle tissue of AA and CF breeds; however, the EAA/NEAA ratio remains unaffected by breed. |

Sci., 2021, 66(3):78-86 | DOI: 10.17221/231/2020-CJAS In the last decade, the pig sector in Slovakia has been changing continuously, especially in regard to the proportion of pigs coming from foreign breeding programs. These changes lead to changes in carcass characteristics and to a change in the distribution of carcasses within the classes of the Union scale for the classification of pig carcasses. Therefore, this study was undertaken to analyse the actual state of pig carcass classification in the Slovak Republic and to investigate the needs of updating the equations for the classification methods and possible steps to improve the quality of pork produced within the country. Results showed that an increased number of pig carcasses originated from Danish breeding program, and also from the Czech Republic and Poland. When compared to carcasses in 2009, these changes resulted in lower backfat thickness and higher muscle thickness of the longissimus dorsi muscle and thus higher lean meat content of carcasses. This results in a higher number of carcasses classified in the S and E classes. Slight differences between two instrumental classification methods were calculated. The study showed the need for updating the equations for classification methods, and also suggested possible further steps in order to improve the quality of pork. |
Effect of thermal management on vitellogenesis and maturation in indoor-reared pikeperch (Sander lucioperca)Original PaperUro¹ Ljubobratoviæ, László Ardó, Georgina Fazekas, Zoltán Horváth, Jovanka LukiæCzech J. Anim. Sci., 2024, 69(1):18-28 | DOI: 10.17221/136/2023-CJAS The present study aimed to assess the effects of thermal manipulation on the vitellogenesis and final oocyte maturation (FOM) in pikeperch. Two groups of fish were stocked in two separate tanks of the climate chamber. One group was stocked at 10-month age (THERMAL), while the other was continually kept under stable photothermal conditions until 19-month-age (CONTROL) and then transferred to a climate chamber. The progress of vitellogenesis was assessed via evaluation of the oocyte diameter at the mid- and late-autumn phases, and the gonadosomatic index (GSI) at the late-autumn phase. Finally, females from the CONTROL group were hormonally stimulated before (WARMING) and after (STABLE) increase of the temperature from wintering to spawning, and the FOM progress, ovulation and plasma levels of 17 α, 20β dihydroxy progesterone (DHP) were assessed. Significantly larger oocytes at the mid-autumn phase (878.8 ± 40.1 μm vs 836.5 ± 46.5 μm) as well as the GSI at the end of the autumn phase (10.5 ± 1.7% vs 7.6 ± 1.1%) were noticed in THERMAL fish. Significantly faster FOM was seen in fish under the WARMING post-stimulation regime, and these fish had higher DHP levels at the moment of hormonal stimulation (5.4 ± 1.4 ng/ml vs 3.8 ± 1.2 ng/ml). According to the obtained results, it appears that photothermal induction of fish at a younger stage might have a positive impact on the first spawning, while the WARMING thermal regime seems to be more efficient in stimulating the FOM in fish upon first wintering. |
Application strategy for sustainable livestock production with farm animal algorithms in response to climate change up to 2050: A reviewReviewSang-O ParkCzech J. Anim. Sci., 2022, 67(11):425-441 | DOI: 10.17221/172/2022-CJAS Global warming caused by climate change can increase heat stress and greenhouse gas (GHG) emissions, leading to food problems and livestock crises. Thus, pre-emptive responses are required to mitigate the food problems and livestock crises. The potential of a livestock crisis caused by global warming highlights the need for sustainable livestock production in response to climate change using a farm animal algorithm in order to address the population increase and avoid food problems in the future. In particular, the demand for animal-based foods has increased. Such a climate change threatens the livestock environment, production, reproductive efficiency, animal behaviour and welfare, while increasing the heat stress, livestock malodours, and GHG emissions. For these reasons, it is necessary to understand the concurrent mechanisms related to these effects of global warming, animal nutrition, animal feeding and management, animal heat stress and in ovo injection, and carbon neutral livestock. Climate-smart livestock systems are being implemented to overcome the livestock crisis caused by climate change and to maintain sustainable livestock production. This review emphasises the importance of sustainable livestock production using farm animal algorithms in response to a future livestock crisis caused by climate change in 2050. |

Understanding the genetic variability of fish stocks is crucial to any animal breeding program. The aim of this study was to perform a meta-analysis of genetic parameters that considers the variability between studies for economically important traits in the Nile tilapia (Oreochromis niloticus) production. The final dataset from literature review contained 226 estimates of heritability and 53 estimates of genetic correlation across 37 peer-reviewed articles published between 2002 and 2022. Weighted genetic parameter estimates were obtained by fitting a random effect model for all traits studied. Heterogeneity across studies were assessed using the Q statistic and I2 index. Heritability estimates ranged from low to moderate magnitude, indicating that a greater response to selection is expected for body length (0.37), body weight (0.29), body width (0.28), and daily weight gain (0.28), compared to the other traits, body height (0.25) and fillet yield (0.19). Genetic correlations ranged from moderate to high magnitude (0.47 to 0.94), which suggests that part of expression of traits is regulated by the same gene groups. The meta-analysis study provided reliable genetic parameter estimates and these findings may be served as the baseline reference for new Nile tilapia (Oreochromis niloticus) breeding programs. |

Sci., 2024, 69(2):48-58 This study aimed to evaluate the effects of whole-plant corn silage (WCS) on growth performance, nutrient availability and intestinal development in growing-finishing pigs. A total of 32 barrows (33.1 ± 3.49 kg) were randomly allocated to four treatments. Control was the basal diet, and the low, medium, and high treatments were basal diets substituted with WCS (air-dry basis) at 5.0%, 7.5%, and 10.0% from day 1 to 42, and 10.0%, 12.5%, and 15.0% from day 43 to 98, respectively. The results showed that the average daily feed intake (ADFI), feed/gain (F/G), and crypt depth (CD) increased linearly (P < 0.05) with the increasing of WCS. However, the final body weight, average daily gain (ADG), biological value (BV), net protein utilization (NPU), dry matter, crude protein (CP), ether extract, crude fibre (CF), and gross energy decreased linearly (P < 0.05). High treatment significantly deteriorated BW, ADG, NPU, and BV compared to control, low, and medium treatments (P < 0.05). The low and medium treatments showed significantly greater ADFI, apparent digestibility (CP and CF), villus height (VH), VH/CD, relative mRNA and protein expression of occludin, claudin-1 and sodium-glucose cotransporter (SGLT1) than the high treatment (P < 0.05). The highest F/G, CD, relative mRNA and protein expression of cationic amino acid transporter (CAT1) were observed in high treatment (P < 0.05). Our results suggested that WCS can replace 7.5% and 12.5% of diet during growing and finishing periods of pigs, respectively. This is of great significance for reducing feeding costs and alleviating food security crises. |

Sci., 2023, 68(12):469-485 | DOI: 10.17221/152/2023-CJAS Flow cytometry (FC) is a very powerful method for the assessment of spermatozoa quality in multiple livestock species, including bulls. Nowadays, many flow cytometric tests are available for quality detection of bull sperm. These tests are based on fluorochromes (fluorescent tags or dyes) and the use of correct concentrations of fluorochromes is an essential aspect of the successful optimization of experimental methodology. Modern flow cytometers are much more affordable, such that spermatologists are now able to use FC methodology (and even image cytometry) in their studies. The present review aims to give a summary of current flow cytometric methodologies used to assess bull sperm quality by flow cytometry. Namely, we give here the concentrations of fluorochromes used in the studies related to bull sperm quality evaluation by flow cytometry published in the last three years (from 2021 to 2023). Importantly, in the present review, the concentration of fluorochromes is unified and presented using comparable molarity units. Furthermore, the rationale of each flow cytometric methodology for particular fluorescent tests, and the citation for the corresponding original methodology, are given. This review will help spermatologists shorten the time needed for the optimization of their flow cytometry methodology for the assessment of bull sperm quality. |
Effects of age and litter-of-origin on cryopreserved spermatozoa in Sumava ramsOriginal PaperAne¾ka Málková, Martin Ptáèek, Filipp Georgijeviè Savvulidi, Szabolcs Tamás Nagy, Ludìk StádníkCzech J. Sci., 2024, 69(4):129-138 | DOI: 10.17221/32/2024-CJAS This study aimed to evaluate the influence of the internal factors of ram age and litter-of-origin on semen quality during the cryopreservation process in Sumava sheep rams. This breed is included in the protected genetic resources of the Czech Republic. The sires were systematically divided into four groups according to age (under 1.5 years, 1.5 to 2.5 years, 2.5 to 3.5 years, and over 4.5 years) and according to litter frequency (singletons vs twins). Semen was evaluated after equilibration, and after cryopreservation using iSperm® mCASA and flow cytometry. During cryopreservation, there was a significant decrease in total sperm motility by 53.5%, progressive motility by 38%, and cells with intact plasma membrane and acrosome by 47%. Frozen-thawed sperm kinematic parameters showed significant age-related variations, with rams aged 4.5 and older displaying notably higher total and progressive motility (16.2% and 6.24%, respectively). Rams born as twins exhibited 3.77% (P < 0.05) higher progressive motility and 5.5% (P < 0.05) higher total motility compared to those born as singles. The sperm of older rams (> 4.5 years) exhibited higher viability (10.1%) and lower damage to the plasma membrane after freeze-thawing (23.6%), (P < 0.05). Twins showed significantly higher sperm viability (4.98%, P < 0.05) than singletons. These rams produce a larger quantity of higher-quality insemination doses after cryopreservation. For Sumava rams, in particular, broadening the sire selection base helps to select suitable rams for breeding. As the sire ages, his genetic value within the production herd may decline with time. This contrasts with genetic resource protection, where the aim is to preserve and store as many high-quality semen samples as possible. |

Sci., 2024, 69(1):1-10 | DOI: 10.17221/156/2023-CJAS Dairy cows enter the negative energy balance in the early post-partum period, which negatively affects milk yield, health status and reproduction. This study aimed to determine if milk yield (MY) and fat/protein (F/P) ratio could be used as reliable indicators for predicting the quality of aspirated oocytes and their further use for in vitro fertilisation. A secondary goal was to compare different periods before oocyte aspiration to determine which period had the most significant impact on the oocyte development in donor cows. All tested Holstein cows were at their first lactation (n = 68). The first ultrasound-guided follicular aspiration in donor cows was done on average at 58.7 days in milk (DIM). Usually, a second aspiration followed a week later if no problems were detected. In total, 102 aspirations were performed. Three model equations with different F/P ratio distributions and variants of MY production were tested in statistical evaluation. On average, 3.6 oocytes were retrieved per donor per aspiration, with a minimum of 1 and a maximum of 12. The maturation rate was 62.2%, with a high probability of reaching metaphase II (90.45%). The highest number of oocytes was obtained from low-yielding cows with a low F/P ratio in all evaluated periods (P < 0.01). The quality and expansion of cumulus-oocyte complexes were unaffected by the metabolic status. However, a tendency for better quality oocytes in donors with lower MY and F/P ratios was observed in all models. Our preliminary results showed that the prediction of oocyte quality in a homogeneous group of the first-lactation cows was related to the chosen data acquisition model. Data about milk yield and F/P ratio obtained from on-farm systems provide a valuable source of information for the selection of oocyte donors. |

Thus, the primary objective of this study was to investigate the effects of dietary housefly maggot extract (HME) as the biofunctional material for antibiotic replacement in laying hens. The biofunctional properties of HME on egg production, egg quality, blood biomarkers of immune function, and microbiota were evaluated over a period of 40 to 50 weeks in laying hens under a digital livestock system. A total of 120 forty-week-old Hyline Brown laying hens were randomly divided into four groups with thirty replicates as follows: no added HME (CON), positive control (PC, 8 ppm of avilamycin), 100 ppm of HME (HME100), and 150 ppm of HME (HME150). Egg production and egg weight increased significantly in the HME150, HME100, PC, and CON groups (P < 0.05), but there were no differences between the HME and PC groups. Egg quality, such as eggshell thickness, eggshell strength, Haugh unit, and albumin height, were significantly improved in the HME and PC groups than in the CON group (P < 0.05). A blood biomarker of immune function, IgG, was significantly higher in the HME and PC groups than the CON group (P < 0.05), but there were no differences between the HME and PC groups. Blood corticosterone and heterophil to lymphocyte ratio were significantly lower in the HME and PC groups than in the CON group (P < 0.05). Microbiota Lactobacillus in the faeces were significantly higher in the HME and PC groups than in the CON group (P < 0.05). The faecal total aerobic bacteria, Escherichia coli, and coliform counts were significantly lower in the HME and PC groups than in the CON group (P < 0.05). Consequently, the HME showed the same significant effects as antibiotics on improving egg production and egg quality in laying hens. These results show that 100 ppm of HME can be used as a biofunctional material for an effective alternative to antibiotics in laying hens under a digital livestock system to improve egg production and quality by stimulating their immune functions and balancing the microbiota populations. |

Sci., 2024, 69(4):155-164 | DOI: 10.17221/22/2024-CJAS Regular replacement of old and problematic queen bees is necessary to maintain the strength and productivity of bee colonies. Beekeepers replace queen bees using queen cells, virgin queens, or mated laying queens. In this study, we explored factors influencing the acceptance of the virgin queens. A comprehensive field study was conducted in the summer of 2023, involving the introduction of 754 virgin queens of different age and 194 queen cells into mating nuclei of various states using three different methods. The article aims to provide recommendations to beekeepers for introducing virgin queens, thereby increasing the frequency of successful requeening. The duration of the colony being queenless has a significant impact on the acceptance of queens. If the colony is prepared in advance, at least five days prior, immature and mature virgin queens will be accepted equally well (60‒77%). Conversely, suppose queen replacement is done without preparation for a colony that has been queenless for one day only. In that case, the result will be significantly lower due to the short queenless period (41%), and the age factor of queen bees will come into play. If queen replacement is done with preparation, the chosen timing within the season will play a significant role, as weather conditions can influence acceptance and mating, varying from 48% to 89%. Suppose a bee colony has long lost its queen and has developed laying worker bees. In that case, there is still a 55% chance of successfully introducing a virgin queen into such a colony without using time-consuming methods. |

A total of 400 one-day-old Isa LabelTM male chicks were used that were distributed in a completely randomized factorial design 2 × 2 + 1 (inclusion of 2% or 4% of corncobs as a source of insoluble fibre IF and citrus pulp as a source of soluble fibre (SF); and a control treatment) with eight replicates of 10 birds each. The addition of IF to diets improved the weight gain and feed conversion of broilers at 7 and 21 days of age. Both fibre sources added to diets resulted in higher relative weights of proventriculus + gizzard, liver, small and large intestine. The diets containing an SF source presented positive effects on the duodenal mucosa. In addition, positive effects on the caecal villus to crypt ratio were observed in broilers fed diets containing different types of fibre. It is concluded that dietary inclusion of IF up to 4% maintains the performance, nutrient metabolism and improves the development of the proventriculus + gizzard in slow-growing broilers. However, for improving the morphological and histomorphological parameters of slow-growing broilers in the starter phase the addition of SF sources is recommended. |

Several strategies have been used to raise the number of piglets to address the challenge of a large litter size. This review provides a rationale for employing the nurse sow system as a biological way of handling large litters, exploring its step-by-step processes and its selected impacts. By exposing these steps, pig farmers will use the information to pinpoint their weak points in the chain and customize the procedures to meet their farm-specific goals for improved productivity. The nurse sow system helps to maximize pre-weaning survival by boosting the overall worth of low birth weight piglets. When piglets weaned per sow/year are used as a performance benchmark, nurse sows weaning double litters become economically advantageous. Extended time in lactation for nurse sows gives them an ample time for their uterus to involute before the next pregnancy. However, nurse sows lose their body reserves reflected in backfat thickness from high milk production. Prolonged confinement in lactation deteriorates the physical condition of nurse sows, resulting in leg ulcers and teat damage. Additionally, piglets are more distressed when taken away from their biological sow to a new sow. |

This indicates that the kind and source of fat in animal diets can significantly impact the accumulation and composition of fatty tissue in the resulting carcass. Hence, feeding (or dietary) manipulation is one of the best and commonly employed management strategies to improve of gut health and function in broiler production. Medium-chain fatty acids (MCFAs) are fatty acids with six to twelve carbon atoms that are prevalent in coconut and palm kernel oils. Much research is being conducted on nutritional approaches to enhance gut health and lower the usage of antibiotics in poultry farming. The use of MCFAs as an antibiotic substitute has been shown to have significant positive effects on broilers’ health and performance due mainly to their short pathways of absorption and oxidation. However, the fatty acid makeup of these feed additions may vary, and the results are sometimes inconsistent. Although supplementing hen diets with MCFAs can lower intermuscular and abdominal fat, the precise mechanics are not entirely understood. The distinct metabolic roles of fatty acids in chickens are linked to both the degree of saturation and the length of the chain. Detailed information on the effect of MCFAs on the growth performance, antimicrobial properties, and fat deposition in broiler chickens are summarised. |
