Optimizing the conventional method of sperm freezing in liquid nitrogen vapour for Wallachian sheep conservation programOriginal PaperFilipp Georgijevic Savvulidi, Martin Ptacek, Anezka Malkova, Jakub Beranek, Ludek StadnikCzech J. Sci., 2021, 66(2):55-64 | DOI: 10.17221/226/2020-CJAS The aim of the present study was to optimize the conventional method of sperm freezing in liquid nitrogen (LN2) vapour for successful cryopreservation of Wallachian ram sperm, the genetic resources of the Czech Republic. Sperm in straws were frozen using the conventional freezing method via a static exposure of sperm doses to LN2 vapour, or by four different modified freezing methods. Under modified freezing, straws were frozen by a discontinuous, time-dependent decremental change in the distance between the straws and the surface of LN2. The viability of sperm was evaluated by flow cytometry after sperm equilibration, and immediately after thawing. Besides the observed inter-sire and daily variation, the obtained results suggest the methodological weakness of the conventional freezing method via the static exposure of sperm doses to LN2 vapour. With the use of the optimized freezing procedure, all parameters of thawed sperm were significantly (P < 0.05) improved in comparison with the conventional method: percentage of thawed sperm viability increased up to 48.3%, percentage of sperm with plasma membrane damage after thawing decreased to 6.58%, percentage of sperm with acrosome damage decreased to 24.4%, and percentage of sperm with deteriorated mitochondrial activity decreased to 6.28%. In conclusion, our results suggest that an optimized freezing procedure should be routinely used instead of the conventional method to cryopreserve Wallachian ram sperm. |

Sci., 2021, 66(5):137-148 | DOI: 10.17221/274/2020-CJAS The aim of this study was to monitor the Czech Aberdeen Angus population for backfat thickness (BT), musculus longissimus lumborum et thoracis (MUSCLE) and live weight (LW) in relation to environmental and genetic conditions. In this study, we also wanted to display the potential for future inclusion of these parameters into national monitoring. Parameters of BT, MUSCLE and LW were measured at 120, 210, and 365 days of age in 769 Aberdeen Angus calves (417 bulls and 352 heifers) on seven farms. Statistical evaluation was performed in the SAS v9.3 software. The bulls achieved significantly higher (P < 0.01) weights and larger MUSCLE compared to the heifers, although the heifers had higher BT. The age of the dam at calving significantly influenced the growth ability of tested calves, but it did not influence BT nor MUSCLE. The nested effect of the year-season, farm and sire bulls (or genetic background) significantly affected LW, BT and MUSCLE of tested calves. The use of the same breeding bulls on different farms resulted in significant differences in growth parameters. Our results displayed the potential of nationwide monitoring of MUSCLE and BT of beef breeds, as official parameters of the beef performance testing in the Czech Republic. The inclusion of these globally used traits in the Czech breeding system would allow to predict breeding values for the Czech beef population. This would lead not only to general improvement of MUSCLE in the population, but also to the production of better breeding animals and slaughter animals with higher value. |
Effects of digestible amino acid based formulation of low protein broiler diets supplemented with valine, isoleucine and arginine on performance and protein efficiencyOriginal PaperNeşe Nuray Toprak, Ismail Yavaş, Ali Anil Çenesiz, Necmettin Ceylan, Ibrahim ÇiftciCzech J. Sci., 2021, 66(5):168-178 | DOI: 10.17221/293/2020-CJAS The aim of the present study was to investigate the effect of digestible amino acid (DAA) based formulation strategy, and l-valine (l-Val), l-isoleucine (l-Ile) and l-arginine (l-Arg) supplementation to reduce the crude protein (CP) level of broiler diets on performance, carcass characteristics and protein efficiency ratio by comparing with the control diet formulated on total amino acid base. A total of 792 one-day-old Ross 308 broiler chicks were divided into 48 floor pens, with 24 pens containing 16 chicks and 24 pens containing 17 chicks. The experiment was organized in a completely randomized block design with four dietary treatments as follows; T1: formulated to meet minimum both total amino acid and DAA requirements without using l-Val, l-Ile, and l-Arg, T2: formulated to meet DAA requirements without using l-Val, l-Ile and l-Arg, T3: formulated to meet DAA requirements by using l-Val alone, T4: formulated to meet DAA requirements by using l-Val, l-Arg, and l-Ile. Each treatment had 12 replicates. Neither l-Val (T3) nor l-Val, l-Ile, and l-Arg supplemented (T4) low CP dietary treatments had any negative impact on growth performance, feed conversion ratio during experimental periods, as well as carcass parameters of broilers. Indeed, CP reduction by the addition of l-Val alone or l-Val, l-Ile, and l-Arg together even resulted in a significant improvement in protein efficiency ratio compared to T1 and T2 treatments. Supplementation of l-Val either alone or along with l-Ile and l-Arg to diets formulated based on DAA not only decreased dietary CP but also soybean meal inclusion. It can be concluded that maintained growth performance, and even improved protein utilization can be achieved due to the DAA based formulation strategy and supplementing broiler diets with l-Val alone or together with l-Ile and l-Arg. |
Evaluation of genetic diversity of subdivided genealogical groups in Lithuanian Trakehner horse population using immunogenetic toolsOriginal PaperAlma Račkauskaité, Ruta Šveistiené, Violeta Razmaité, Virginija JatkauskienéCzech J. Anim. Sci., 2021, 66(6):200-210 | DOI: 10.17221/118/2020-CJAS The objective of this study was to evaluate the inter-breed genetic diversity of Lithuanian Trakehner horses. The current population in Lithuania numbers 1 039 Trakehner horses. The study included the immunogenetic data analysis of 316 blood samples. Pedigree was traced back to 7-11 generations. The horses were assigned to genealogical clusters originating from East Prussian Trakehner Stud, Thoroughbred and Arab sires. The first two clusters were subdivided into nine-horse groups according to the most important ancestors. The evaluation of genetic diversity was based on six blood groups and five protein polymorphism systems. The differences between genealogical groups were confirmed by principal component analysis and applied cluster analysis. According to the allelic frequency, the Trakehner population changed very slightly through a 30-year period, and only one rare (0.013) allele TfRR (frequent in other Trakehner populations), has been newly found. The analysis of genetic polymorphism systems based on a very low rate of alleles Qabc (0.093) and Ddkl (0.046), which are typical of other populations, indicated the exclusivity of the Trakehner population in Lithuania. The average expected heterozygosity by blood protein polymorphism and blood groups were 0.409 and 0.441, respectively. The genetic clustering diverged by observed heterozygosity of blood groups and by pedigree data in Pilger through Egoist, Bay Ronald through Dark Ronald and Dampfross through Hyperion subgroups. The determined distinctiveness of subdivided Trakehner horse groups suggests reconsidering the breeding strategies and conservation programme of Trakehner horses. Breeding and selection of subdivided sire lines could be among the appropriate solutions for the maintenance and extension of the genealogical structure. |

Sci., 2022, 67(1):27-38 | DOI: 10.17221/136/2021-CJAS This study was conducted to investigate the effect of Lactobacillus zeae LB2 on the gut microbiota in piglets infected with enterotoxigenic Escherichia coli (ETEC). Thirty-two healthy 7-day-old piglets were randomly divided into four treatment groups: control group (basal diet), LB2 group (supplemented with 1 × 108 CFU/pig/day L. zeae LB2), ETEC group (infected with 1 × 1010 CFU/pig/day ETEC) and ETEC+LB2 group (LB2 supplementation + ETEC infection). Intestinal contents were collected for DNA extraction and Illumina sequencing. Significant result was observed for alpha diversity in the four intestinal sections, and both ETEC infection and LB2 supplementation showed a higher Chao1 alpha diversity. At the phylum level, Firmicutes and Bacteroidetes were dominant in the healthy piglets, while Proteobacteria were dominant in the ETEC-infected piglets. At the genus level, ETEC infection decreased the abundance of Prevotella, Ruminococcaceae, Lactobacillus, Alloprevotella, Flavobacterium, and Sutterella and increased the abundance of Actinobacillus. The LB2 supplementation reduced the abundance of Ruminococcaceae, Actinobacillus, Porphyromonas, and Alloprevotella, and increased the abundance of Prevotella and Lactobacillus. Both ETEC infection and LB2 supplementation affected several functional pathways associated with cellular processes, environmental information processing, genetic information processing, diseases, metabolism, and organismal systems. In summary, ETEC infection induced dysbiosis of the gut microbiome in piglets, while L. zeae supplementation could positively regulate the gut microbiome during ETEC infection. Therefore, L. zeae LB2 may be an ideal probiotic for the prevention or treatment of ETEC infection. |
Effects of dried Portulaca oleracea supplementation to the laying hen diet on productive performance, egg physical traits, fatty acid composition, and cholesterol contentOriginal PaperAntonella Dalle Zotte, Gianluca PranzoCzech J. Sci., 2022, 67(3):114-123 | DOI: 10.17221/9/2022-CJAS Portulaca oleracea is a widespread herbaceous plant particularly rich in polyunsaturated fatty acids (PUFA), antioxidant compounds and characterised by a healthy omega-6/omega-3 ratio. The focus of this research was to evaluate the effects of Portulaca oleracea supplementation to the diet of laying hens on productive performance, egg physical traits, fatty acid composition and cholesterol content. Twenty-six 24-week-old Warren strain layers were randomly assigned to two different groups of 18 and eight birds, respectively: one group received a commercial diet (C) whereas the other group was given the same control diet supplemented with 20% of dried Portulaca oleracea (PO). Hens were fed for 21 days. Feed and water were provided ad libitum. The poor palatability of the PO diet compromised the optimisation of the productive performance, with a significant reduction of the oviposition efficiency (0.69 vs 0.88 for PO and C, respectively; P < 0.05) and egg physical traits. Considering the egg nutritional traits, dietary PO significantly decreased the yolk proportion of saturated fatty acid (43.0% vs 44.1%, P < 0.05), while it increased the content of PUFA (19.4% vs 17.8%, P < 0.001), and within the latter, both omega-6 and omega-3 proportion significantly increased in comparison with C group (16.4% vs 17.6%, P < 0.001 and 1.46% vs 1.80%, P < 0.001, for n-6 and n-3, respectively). This resulted in an improvement of the omega-6/omega-3 ratio (10.4 vs 11.3 for PO and C, respectively; P < 0.05). Egg yolk cholesterol content did not vary between dietary treatments. It could be concluded that the use of Portulaca oleracea for producing omega-3-enriched eggs is feasible once the problem of palatability has been overcome and the energy requirements of the hens have been covered. |
A meta-analysis of heat stress in dairy cattle: The increase in temperature humidity index affects both milk yield and some physiological parametersOriginal PaperErsin Kulaz, Gazel SerCzech J. Anim. Sci., 2022, 67(6):209-217 | DOI: 10.17221/1/2022-CJAS In this study, the relationships of temperature humidity index (THI) with milk yield and some physiological responses in dairy cattle were investigated. Our goal in the meta-analysis was to find the parameter(s) primarily affected under heat stress. A total of 16 studies with the temperature humidity index value higher than 72, which is an important factor in determining the effect of heat stress, were included in the meta-analysis. The variables of interest in the meta-analysis included: milk yield (kg/day), respiratory rate (breaths/min), rectal temperature (°C). In addition to the meta-analysis, principal component analysis (PCA) was also performed. In the meta-analysis, high variation or heterogeneity (I2 > 99%) was determined between the results of the studies. This may depend on many factors (climate, region, number of samples and management etc.). Heterogeneity is desirable in the meta-analysis, because it provides accurate and reliable interpretations of the variances of parameters. Due to high heterogeneity, the results of the studies were combined according to the mixed model. According to the mixed model and PCA results, a linear relationship was determined between the temperature humidity index and these physiological parameters. According to the meta-analysis, at THI > 72, the mean effect size of milk yield was 50%, and the effect sizes of respiratory rate and rectal temperature were approximately 65% and 38%. All three parameters have a significant effect under heat stress (P < 0.000 1). As a result, there is a linear relationship between temperature humidity index, milk yield and physiological parameters. According to the other characteristics, the respiratory rate was determined as the primary response parameter in parallel with the increase in temperature humidity index. |
Dietary brown seaweed and plant polyphenols in hyperprolific sows: Productive performance, blood parameters and antioxidant statusOriginal PaperRaffaella Rossi, Telma Tucci, Alessia Giordano, Pierangelo Moretti, Carlo CorinoCzech J. Sci., 2022, 67(12):493-502 | DOI: 10.17221/98/2022-CJAS This study evaluated the productive performance, blood parameters and antioxidant status of hyperprolific sows fed control diet or diet supplemented with brown seaweed and polyphenol mixture (SPM). Ninety-six farrowing highly prolific sows (Topigs 20) were assigned to two dietary treatments from day 107 ± 2 days of gestation until weaning: control diet and the same diet containing 15 g/day of SPM. Sows fed SPM diet tended to have lower backfat losses (P = 0.06) than control sows. No difference was observed in daily feed intake. No difference in litter birth weight or number of piglets per litter was observed. In sows fed SPM, average daily gain and weaning body weight of piglets were increased (P < 0.05). Haematochemical parameters, haptoglobin and blood total antiradical activity were not affected (P > 0.05) by dietary treatment. The paraoxonase-1 activity was higher (P < 0.05) in SPM sows than in the control. At the subsequent farrowing, the total number of piglets born was higher (P < 0.05) in sows fed SPM than in the control. Overall, these data suggest that dietary plant polyphenols and brown seaweeds improved the weaning weight of piglets and the total number of piglets born at the subsequent farrowing, modulating oxidative stress in lactating highly prolific sows. |

Sci., 2019, 64(1):1-10 | DOI: 10.17221/35/2018-CJAS Verbascoside, the main component of Lippia citriodora extract, is one of the most powerful free radical scavengers exhibiting a wide biological activity. In in vivo study 20 adult New Zealand white rabbit bucks were divided into two homogeneous groups, one control (CON) and one verbascoside-supplemented (0.1%) in feed mixture (EXP) and later in vitro effects of verbascoside on the motility aspects of rabbit spermatozoa were analysed. The spermatozoa concentration, ejaculate volume, spermatozoa motility, progressive motility, distance parameters, velocity parameters and type of spermatozoa movement were negatively affected by Lippia citriodora leaves extract after the first 4 weeks of dietary treatment, till the end of experiment (8 weeks). Four weeks after the suspension of feed additive supplementation, all spermatozoa traits values returned to the normality, and in line with CON group. For in vitro findings, ejaculates from 10 male New Zealand white bucks were collected using an artificial vagina. Then it was diluted in physiological saline solution containing different concentrations of verbascoside at the concentration of 0, 0.0024, 0.0219, 0.157, 120.0 mg/ml (Ctrl, VB1, VB2, VB3, VB4 groups, respectively), using a dilution ratio of 1 : 4. The obtained data proved that verbascoside at the concentration of 0.0024 and 0.0219 mg/ml had no adverse effect on spermatozoa. Additionally, we found that verbascoside at higher concentrations (0.157 and 120.0 mg/ml) significantly altered all the motility parameters analysed in the experiment. In conclusion a possible negative effect of verbascoside supplementation into feed mixture (0.1%) on semen quality parameters in rabbit bucks as well as in vitro can be stated, obviously considering that target organs of antioxidant activities of phenylpropanoid glycosides are various. In addition it has to be emphasized that the extract showed a reversible action, since the semen traits of treated animals returned to the normality after the dietary administration period. |
Principal component analysis of conformation traits in Hungarian Simmental cowsOriginal PaperEvelin Török, István Komlósi, Béla Béri, Imre Füller, Barnabás Vágó, János PostaCzech J. Anim. Sci., 2021, 66(2):39-45 | DOI: 10.17221/155/2020-CJAS The aim of the current research was to analyze the linear type traits of Hungarian Simmental dual-purpose cows scored in the first lactation using principal component analysis and cluster analysis. Data collected by the Association of Hungarian Simmental Breeders were studied during the work. The filtered database contained the results of 8 868 cows, born after 1997. From the evaluation of main conformation traits, the highest correlations (r = 0.35, P < 0.05) were found between mammary system and feet and legs traits. Within linear type traits, the highest correlation was observed between rump length and rump width (r = 0.81, P < 0.05). Using the principal component analysis, main conformation traits were combined into groups. There were three factors having 84.5 as total variance ratio after varimax rotation. Cluster analysis verified the results of the principal component analysis as most of the trait groups were similar. The strongest relationship was observed between feet and legs and mammary system (main conformation traits) and between rump length and rump width (linear type traits). |

Sci., 2021, 66(9):339-348 | DOI: 10.17221/86/2021-CJAS Cadmium (Cd) is a toxic heavy metal, discharged into environments naturally or anthropogenically, where industrial and agricultural revolutions have led to serious Cd pollution into the environment. Poultry feed may be contaminated by Cd due to its presence in concentrates and supplements, and also in the machinery and equipment used in feed production and processing. After absorption in the digestive system, Cd largely accumulates in kidneys, liver, bones and other organs and causes irreversible damage to these organs. Higher doses of Cd in the poultry diet reduced feed intake, body weight, bone mineral density, egg production and eggshell quality. Selenium (Se) is an essential micronutrient and important component of a variety of antioxidant enzymes, with the ability to antagonize metal toxicity including Cd. Hence, in the current review, we try to summarize recent information about the toxicological effects of Cd and provide updated information about the role of Se as an effective strategy in reducing Cd toxicity in poultry. |

Sci., 2021, 66(9):381-390 | DOI: 10.17221/54/2021-CJAS The aim of this study was to examine the effects of ozonation on the water quality, and growth, blood biochemistry, antioxidant capacity and survival of pikeperch (Sander lucioperca) reared in a recirculation aquaculture system for 30 weeks. A group without ozone treatment was used as a control. The ozone application led to a significant reduction of the water chemical oxygen demand, biological oxygen demand and unsuspended solids concentration. The results revealed that an ozone treatment as a water treatment method has a positive influence on the intensive culture of pikeperch ensuring a higher survival rate (77%) compared to the non-treated control group (67.2%). Moreover, the ozonation prevented fin damage to a large extent and reduced the prevalence of an Ichthyophthirius multifiliis infection. Furthermore, the ozone application led to a reduction in the thiobarbituric acid reactive substance level and enhanced the superoxide dismutase activity in the fish gills. However, the effect of ozonation was null on the plasma biochemical parameters. Overall, these findings suggest that an ozone treatment, using adequate technological equipment to destroy the residual ozone, improves the water quality and protects pikeperch against any possible infection and fin damage in a recirculation aquaculture system. |
Comparison of starch digestibility methods for extruded wheat grains (Triticum aestivum L.)Original PaperDana Homolková, Vladimír Plachý, Anna Maňourová, Michal Kaválek, Václav Dvořáček, Boris Hučko, Milan Marounek, Ivo DoskočilCzech J. Sci., 2021, 66(10):420-427 | DOI: 10.17221/56/2021-CJAS This study compared different methods of determining starch digestibility (in vivo vs in vitro) in wheat grains and evaluated the influence of extrusion on digestibility. In vivo starch digestibility was determined in broiler chickens by calculating the residual starch content in their ilea and the digestibility using a chromium oxide indicator. In vitro digestibility was examined using pepsin and pancreatin. During in vivo testing, the highest digestibility coefficient (DC) was achieved by the Bonanza variety in its extruded form (91.19 ± 0.40%). In contrast, the lowest DC was achieved by the Tobak variety in its non-extruded form (81.45 ± 1.92%). Generally, a higher DC was observed in vivo for extruded forms of wheat. During in vitro testing, the highest DC was achieved by the Stefii variety in its non-extruded form (96.10 ± 0.55%), whereas the lowest DC was observed in the Yetti variety in its extruded form (49.72 ± 0.41%). Overall, the in vitro experiments did not exhibit significant differences between extruded and non-extruded forms of wheat. Linear regression analysis showed a strong relationship (r2 = 0.860; 85.98%) between in vivo- and in vitro-derived DC values in all wheat varieties, both in extruded and non-extruded forms. The study showed that in vivo testing is a suitable method for the determination and control of starch levels in extruded materials. However, despite the accuracy of this technique, it is also very demanding in terms of time, space, equipment, and methodological knowledge. Therefore, based on the strong correlation between the in vivo and in vitro assays, we recommend in vitro digestibility testing as a preferable alternative. |
Protection of bovine mammary epithelial cells from hydrogen peroxide-induced oxidative cell damage by seleniumOriginal PaperYong Mei Guo, Jian Gong, Ya Guang Zheng, Bin Lin Shi, Xiao Yu Guo, Su Mei YanCzech J. Sci., 2018, 63(3):94-102 | DOI: 10.17221/76/2017-CJAS The uncontrolled release of arachidonic acid (ARA) and its metabolism by lipoxygenase (LOX) pathway can induce and aggravate cellular oxidative stress. Selenium (Se) is an integral part of some antioxidative selenoproteins and may protect cells from oxidative damage by modulating ARA release and metabolism. The present study aimed to investigate the protective response of Se against hydrogen peroxide (H2O2)-induced oxidative damage in bovine mammary epithelial cells (BMECs). The BMECs were incubated for 24 h in serum-free medium and then divided into four groups randomly. The cells in groups 1 and 2 were subsequently incubated for 30 h in serum-free medium containing 0 (control) and 50 nM Se (Se treatment group). The cells in groups 3 and 4 were incubated for 24 h in serum-free medium containing 0 and 50 nM Se, and then treated with 600 μM H2O2 for 6 h (H2O2 damage group and Se prevention group). The results showed that Se attenuated the H2O2-induced production of reactive oxygen species and the decrease of antioxidative enzymes as glutathione peroxidase (GPX), thioredoxin reductase (TrxR), selenoprotein P (SelP), superoxide dismutase, and catalase in BMECs. The preventive effects of Se on the decrease of selenoprotein activity were demonstrated further by the increase of mRNA expression for GPX1, TrxR1, and SelP, and protein expression for GPX1 and TrxR1. Pretreatment of cells with Se inhibited the H2O2-induced increase of mRNA expressions and activities for cytosolic phospholipase A2 and 5-lipoxygenase, ARA release, and 15-hydroperoxyeicosatetraenoic acid production. Se also blocked the H2O2-induced activation of p38 mitogen-activated protein kinase (MAPK) and c-Jun N-terminal kinase but not that of extracellular signal-regulated kinase. These results suggested that Se may protect BMECs against H2O2-induced oxidative damage by increasing selenoproteins synthesis, inhibiting MAPK pathway, and then decreasing ARA release and its metabolism by LOX pathway. |

Curcumin (CUR) has become a substance of scientific interest for its free radical-quenching abilities, which could enhance the post-thaw quality of male gametes. This study assessed the effects of CUR on the post-thaw vitality and selected oxidative stress markers of bovine spermatozoa. Thirty ejaculates collected from 10 breeding bulls were divided into two aliquots and cryopreserved in the absence (control) or presence of CUR (50 μmol/l). Immediately before use, the control or experimental straws were thawed at 37°C for 20 s. CUR administration led to a significantly higher preservation of spermatozoa motion (P < 0.001) as well as membrane (P < 0.05) and acrosomal (P < 0.01) integrity in comparison with the control. Moreover, spermatozoa exposed to CUR exhibited a significantly higher mitochondrial activity (P < 0.001). Significantly decreased amounts of reactive oxygen species (P < 0.01) and superoxide (P < 0.001) were detected following CUR supplementation. Finally, a significant decrease of oxidative damage to proteins (P < 0.01), lipids (P < 0.001), and DNA (P < 0.05) was recorded in samples to which CUR was administered in comparison to the control. In this study, CUR proved to act as an efficient antioxidant molecule offering protection to male gametes against oxidative damage during cryopreservation. |
Analysis of raw cow milk quality according to free fatty acid contents in the Czech RepublicO. Hanuš, J. Vegricht, J. Frelich, A. Macek, M. Bjelka, F. Louda, L. JanůCzech J. Anim. Sci., 2008, 53(1):17-30 | DOI: 10.17221/2717-CJAS The concentration (c) of free fatty acids (FFAs) in milk is an indicator of dairy cow nutrition, milk straining, its bacterial contamination and storage quality. High FFA concentrations (cs) caused by lipolysis can damage the quality properties of milk products. Therefore the FFA content is introduced thanks to an increase in the efficiency of modern analytical methods as a milk quality indicator and as an indicator for its price as well. The goal of this paper was to analyse the FFA relations to the other milk quality indicators. The data set (n = 11 586) was evaluated by regression methods. In November and December the respective FFA means were 0.614 ± 0.458 and 0.835 ± 0.491 mmol/100 g with a relatively high variability of 74.6 and 58.8%. The frequency of unsatisfactory FFA values (> 1.3) was 7.51 and 13.93%. Casein content (r = -0.17; P < 0.01) and crude protein content (r = -0.12; P < 0.01) were related more closely with FFA c. The FFAs can increase by 0.066 mmol/100 g with casein decrease by 0.10%. The FFAs in milk fat can slightly increase by the supply of energy to dairy cows (protein and casein decrease) and rise with the deteriorating health state of mammary gland (lactose, r = -0.14; P < 0.01) as well. The somatic cell count correlated with FFAs more weakly (r = 0.07; P < 0.05), similarly like the total mesophilic bacteria count (r = 0.11; P < 0.01), relatively more closely the psychrotrophic bacteria count (r = 0.27; P < 0.05). The deterioration of almost all hygienic indicators signified an FFA c increase. The urea content correlated with FFAs weakly (r = -0.08; P < 0.05) and the fat content imperceptibly as a component of similar substance like FFAs. The mechanical milk stress led to FFA liberation from fat esters proportionally to the intervention intensity (P < 0.001). Even a relatively small mechanical stress caused by mixing comparable to the current milking technology, milk transport and storage increased the FFA c of milk fat from 1.11 ± 0.19 to 1.80 ± 0.40 mmol/100 g (P < 0.05). The highest experimental stress up to 6.88 ± 0.55 mmol/100 g (P < 0.001). |

The digestibility analysis was carried out by two in vivo methods, total faeces collection (TFC) and using lignin as a marker (ADL). Animals were divided into 3 groups: control group (C, without supplementation), experimental group 1 (E1, feed rations + 200 g of DGP) and experimental group 2 (E2, feed rations + 400 g of DGP). In animals, no health problems were detected during the trial. Of the blood serum indicators, only the concentrations of potassium (increase in E2 group compared to C group) and alanine aminotransferase (decrease in E2 group in comparison with E1 and C group) were affected (P < 0.05). The ADL method resulted in underestimated digestibility coefficients due to low recovery rates of lignin (less than 90%) in C group and E1 group. According to TFC, in E1 group higher digestibility coefficients were detected for DM, OM and CP (P > 0.05) compared to C group. However, in E2 group lower digestibility of all the studied nutrients was found (P > 0.05) in comparison with C group and E1 group. These results suggest that DGP could be used in horse diets up to 200 g without negative effect on their health and for a possible digestibility improvement of some nutrients. |

Sci., 2020, 65(9):337-345 | DOI: 10.17221/192/2020-CJAS The aim of the study was to evaluate the effect of crossing Mecklenburger Schecke (MS) males with the maternal commercial line of HYLA rabbits on the growth performance, mortality and body and carcass components of fattened rabbits under intensive farming conditions. The control (C) group consisted of albino crossbred rabbits of the HYLA combination AB sires × CD dams. The tested (T) group (MS sires × HYLA CD dams) consisted of rabbit crosses between the MS sires and the same dam line as in the C group. Rabbits of both groups were raised and fattened under identical nutritional and management conditions. The crossing of MS males with the maternal line of HYLA rabbits led to the acceptable growth performance of rabbits in the T group. The higher slaughter weight of rabbits in the T group as compared to rabbits in the C group (P < 0.01) as associated with the higher weight of hot carcass and higher proportions of fore and intermediate parts of carcass (P < 0.01). On the contrary, HYLA rabbits had a higher proportion of the hind part of carcass (P < 0.001); yields of hind leg meat and musculus longissimus thoracis et lumborum were not influenced by the genotype. However, since higher mortality was found in rabbits of the T group during fattening, it is not possible to recommend the MS breed as a common sire line used under conditions of intensive farming at this moment. Regarding the rabbit sex, slaughtered females showed a higher proportion of liver and a lower dressing percentage associated with the worse carcass compactness compared to males. |

Enterococcus faecium AL41, enterocin M-producing strain (deponed to the Czech Culture Collection of Microorganisms, Brno, Czech Republic - CCM8558) is our isolate previously applied e.g. in broiler rabbits with beneficial effect. In this study it was used in a 11-day experiment with chickens (1-day-old, breed Cobb 500, n = 40) challenged with C. jejuni. Birds had free access to feed and water; they were randomly divided into four groups per 10 chicks each: control (CG), E. faecium CCM8558 (EG1), CCM8558 + C. jejuni CCM6191 (EG2), CCM6191 (EG3). E. faecium CCM8558 (109 CFU/ml, 200 µl) in Ringer solution was administered daily per os to EG1 and EG2 for 7 days (from day 0-1). EG3 and EG2 were infected individually per os (day 4, CCM6191, 108 CFU/ml in Ringer solution, 200 µl). For microbiota evaluation, faecal mixtures (n = 5) were sampled on day 0-1 (1st sampling), on day 7 (2nd sampling), and on day 11 (3rd sampling) (day 7 of CCM8558 application, day 4 post-infection; day 4 of CCM8558 cessation, day 7 post-infection). Five birds from each group were sacrificed. CCM8558 sufficiently colonized chickens. In faeces of EG2 a tendency to reduce Campylobacter spp. (day 7 of application, day 4 of infection) was noted compared to EG2 (day 11, day 4 of cessation, day 7 post-infection; difference 1.21 log cycles), while in EG3 CCM6191 strain was not reduced. Phagocytic activity (PA) values were significantly higher in infected groups compared to CG and EG1. A significant increase in PA was also noted in EG2 and EG3 at the end of experiment compared to CG or EG1. The strain additive did not evoke oxidative stress. Biochemical parameters were influenced to the reference levels. |

Subconfluence BMEC were randomly divided into four groups with six replicates: the control group (incubated in serum-free medium without RA or H2O2 for 30 h), H2O2 group (pre-incubated for 24 h without RA, then for another 6 h with 600 μM H2O2), RA group (incubated with 1 mg/ml RA for 30 h without H2O2), and RA + H2O2 group (RA prevention group, pre-incubated with 1 mg/ml RA for 24 h and then for another 6 h with 600 mM H2O2). The results showed that the H2O2 treatment significantly decreased several measured traits, including the cell viability, glutathione peroxidase (GPX) and thioredoxin reductase (TRXR) activities, selenoprotein P (SELP) content, catalase and superoxide dismutase activities, total antioxidant capacity, and GPX1, TRXR1, and SELP gene expression, as well as GPX1 and TRXR1 protein expression. H2O2 treatment also increased the malondialdehyde and reactive oxygen species contents and induced a marked increase of several measured traits, including the arachidonic acid (ARA) concentration, cytosolic phospholipase A2 and 5-lipoxygenase gene expression and activity, and 15-hydroxy twenty-four arachidonic acid and hydroxy peroxide tetracosenic arachidonic acid contents. RA pre-treatment prevented corresponding increases in parameters related to ARA metabolism and increased the activity of TRXR. Moreover, RA pre-treatment attenuated the phosphorylation levels of p38 mitogen-activated protein kinase (MAPK) and c-Jun N-terminal kinase and effectively decreased the ARA content. These results suggest that RA protected BMEC from oxidative stress by elevating TRXR activity, which inhibited the MAPK signaling pathway and led to a decreased concentration of ARA. |
Relationship between abnormal spermatozoa and seminal plasma free amino acids in boarsJ. Čeřovský, S. Frydrychová, A. Lustyková, M. RozkotCzech J. Anim. Sci., 2007, 52(2):44-49 | DOI: 10.17221/2346-CJAS The objective of this study was to provide some data on concentrations of seminal plasma free amino acids (FAAs) in order to compare these values with different total contents of morphologically abnormal spermatozoa (MAS) in the semen of 37 experimental adult boars. The boars were kept in the same housing, feeding and breeding conditions. Immediately after collection, semen gel free volume, sperm concentration and total MAS were determined microscopically. The boars were divided into two groups (A - n = 24, B - n = 13) according to the significantly different total MAS content (x: A = 9.46 ± 4.95, B = 46.00 ± 14.54%, P < 0.01). Deproteinized seminal plasma samples (supernatant) were analysed for concentrations of 13 FAAs: Tau, Asp, Thr, Ser, Glu, Gly, Ala, Val, Met, Ile, Leu, Tyr and Lys by liquid chromatography (AAA 339 M analyser). A highly significant correlation was determined between total MAS and total FAA content in all experimental boars (n = 37, r = -0.60, P < 0.01). Glutamic acid was a predominant FAA in seminal plasma in both groups of boars (x: A = 28.49 vs. B = 17.60μM/100 ml) but its concentration was significantly higher in group A (P < 0.01). The proportion (%) of glutamic acid concentration in the total content of FAAs in both groups of boars was nearly equal (A = 38.1 vs. B = 38.9%, P > 0.05) as well as the proportion index of the individual FAAs calculated from glutamic acid (= 100%). The differences in the particular FAAs recorded between group A and group B were statistically significant for 11 out of the 13 FAAs observed (P < 0.05 and P < 0.01) in favour of group A as well as in the total calculated content of FAAs per boar (x: 74.70 vs. 45.23μM/100 ml, P < 0.01). A significantly negative relationship between the MAS content and the concentration of seminal plasma FAAs (r = -0.60, P < 0.01) is the main result of this study with regard to the markers for potential boar semen fertility estimation. |

RE-NUM-OR was written using Python (Ver. 2.7.13) 64 byte programming language and compiled with PyInstaller-3.2.1 software which is a set of utilities for freezing Python scripts into executable. RE-NUM-OR not only runs on 32 or 64 byte MS Windows but also runs on 64 byte GNU/Linux. The program has new, practical, and simple interface and the user does not need to create a parameter file for running the program and .txt, .xlsx or .et extension files can be used as input files directly. Output files (.txt, .xlsx, .et, .dat) can be obtained. For animal, sire and dam IDs, it can read all standard characters (ASCII codes from 32 to 126) in input files. The program supports both dot (.) and comma (,) for numerical data. Pedigree lines for parents do not need to be arranged prior to their progeny. If pedigree lines for parents follow their progeny, the program can detect this and it can reorder the animals as pedigree lines placing parents prior to their progeny. Another feature of this software is that it has a pop-up window with error notification. Also, it supports repeated observations. Sci., 2018, 63(10):408-418 | DOI: 10.17221/44/2018-CJAS The objective of this study was to calculate economic weights for ten current breeding objective traits and for four new traits characterising claw health and feed efficiency in Czech Holstein cattle and to investigate the impact of different selection indices on the genetic responses for these traits. Economic weights were estimated using a bio-economic model, while applying actual (2017) and predicted (2025) production and economic circumstances. For the actual situation, the economic weights of claw disease incidence were -100.1 € per case, and those of daily residual feed intake in cows, breeding heifers, and fattened animals were -79.37, -37.16, and -6.33 €/kg dry matter intake per day, respectively. In the predicted situation, the marginal economic weights for claw disease and feed efficiency traits increased on average by 38% and 20%, respectively. The new traits, claw disease incidence and daily residual feed intake, were gradually added to the 17 current Holstein selection index traits to improve the new traits. Constructing a comprehensive index with 21 traits and applying the general principles of the selection index theory, a favourable annual genetic selection response was obtained for the new traits (-0.008 cases of claw disease incidence and -0.006 kg of daily residual feed intake across all cattle categories), keeping the annual selection response of the most important current breeding objective traits at a satisfactory level (e.g., 73 kg of milk yield per lactation, 0.016% of milk fat). Claw health and feed efficiency should be defined as new breeding objectives and new selection index traits of local dairy population. |
Multi-breed herd approach to detect breed differences in composition and fatty acid profile of cow milkOriginal PaperCarmen L. Sci., 2019, 64(1):11-16 | DOI: 10.17221/18/2018-CJAS The objective of the present study was to estimate the effect of breed on milk fatty acid (FA) composition of dairy (Brown Swiss, Holstein-Friesian, and Jersey) and dual-purpose cows (Simmental and Alpine Grey) in multi-breed herds. Information on individual milk samples was collected during routine cow milk testing between 2011 and 2014, and consisted of 285 606 observations from 17 445 cows in 617 herds. Fixed effects included in the mixed model were breed, parity, stage of lactation and the interaction between parity and stage of lactation, and random effects were cow, herd-test-date and residual. Contrast estimates for the studied traits were used to compare specific sets of breeds. Holstein-Friesian produced more milk than the other cattle breeds, with the greatest trans FA and C18:1 and the lowest C18:0 content. Comparison between the specialised dairy vs the dual-purpose breeds highlighted significant differences for all traits except for polyunsaturated FA and trans FA content. Specialised dairy breeds had greater milk saturated FA, short-chain FA, medium-chain FA, C14:0 and C16:0 content, and dual-purpose breeds produced milk with greater content of monounsaturated FA, long-chain FA, C18:0 and C18:1. Results demonstrated that, although specialised dairy produced more milk than dual-purpose breeds, milk FA profile of the latter was more favourable from a human nutrition point of view. |
Comparison of different semen extenders and cryoprotectant agents to enhance cryopreservation of rabbit spermatozoaOriginal PaperPaula Domingo, Maite Olaciregui, Noelia González, Ignacio De Blas, Lydia GilCzech J. Sci., 2019, 64(2):59-66 | DOI: 10.17221/53/2018-CJAS The purpose of this research was to find a suitable protocol to enhance frozen rabbit sperm preservation analysing the role that seminal plasma (SP) plays and the effect of different cryoprotectant agents on sperm quality 0 and 2 h after thawing. Sperm samples were pooled and divided in eight fractions. Four of them were diluted with BotuCrio® (extender A), INRA 96® plus 6% glycerol (extender B), 6% N, N-dimethylformamide (extender C) and 6% N-methyl-2-pyrrolidone (extender D), respectively. The other four fractions were centrifuged and the supernatant was discarded in order to eliminate SP. Each sample was then resuspended with extender A, B, C and D. Samples were cooled progressively, loaded into 0.5 ml freezing straws and frozen with liquid nitrogen vapour. Thawing was performed by placing the straws into a bain-marie at 37°C for 21 s. Straws were dried and sperm samples placed into Eppendorf tubes to be analyzed by ISAS software, vitality test, HOS test and acrosome integrity test. The best motility and velocity parameters were obtained by extender A (P < 0.050) even when the motility parameter was compared with previous studies using other diluents. Additionally, sperm quality decreased over incubation time (P < 0.050) and no differences were found in samples processed with or without SP. This research revealed that BotuCrio® could be used for rabbit sperm cryopreservation and moreover the improvement of the cryopreservation process of rabbit sperm due to the demonstration that SP removing is not required. |

However, under cold and/or moderate ambient temperature, these genes might possess different behaviour. The main objective of this study was to evaluate laying performance, immune response, and antioxidant status of native laying hens segregating for naked neck (Na) and frizzle (F) genes under low ambient temperature. Five genotypes were studied: homozygous naked neck (NaNaff), heterozygous naked neck (Nanaff), homozygous frizzle (nanaFF), heterozygous frizzle (nanaFf), and normally feathered (nanaff). The hens were raised under temperature range 22.2-16.7°C. No adverse effect due to ambient temperature was detected in laying performance for naked neck genotypes. Significant decrease in egg weight was recorded in nanaFF genotype compared to the other genotypes leading to significant decrease in egg mass. Significant improvement in shell thickness was associated with Na and F genes. NaNaff genotype had a significantly higher eggshell strength compared to nanaff. Furthermore, Na and F genes improved cellular mediated immune responsiveness, whereas this improvement did not extend to humoral immunity. Birds carrying F gene in homozygous state had a higher total antioxidant activity compared to the remaining genotypes. It could be concluded that the presence of Na and F genes in laying hens raised under low ambient temperature significantly increased shell thickness and, in turn, improved shell strength. Moreover, they greatly enhanced cellular immunity, particularly in heterozygous naked neck status. |

A total of 60 calves were divided into two groups. Group A/B was fed according to a two-phase calf nutrition system: the calves received calf starter "A" (28.5% starch content, 16% neutral detergent fibre (NDF), 9.2% acid detergent fibre (ADF)) in days 7-45 and then calf starter "B" (14.2% starch content, 32.4% NDF, 14.3% ADF) in days 46-70. Calves of group B received calf starter "B" throughout the experimental period. The calves were weaned between 52 and 61 days of life. The data on body weight, weight gain, starter feed intake, blood metabolites and rumen fermentation parameters were collected individually. In the first phase of experiment, the starter feed consumption of A/B group was higher than that of B group (P < 0.05), but it did not manifest in differences in body weight at weaning time. At some of the sampling times, the different composition of starter diets significantly (P < 0.05) influenced the plasma urea, albumin, glucose and triglyceride concentrations. The consumption of diet with different starch and NDF content had no significant effect either on the composition of the rumen microbiota or on the ruminal concentration of volatile fatty acids (VFA) and ammonia. In some cases significant correlations (-0.83 and -0.93; 0.82 and 0.90) were found between the composition of rumen microbiota and ruminal VFA concentration. Feeding a diet of higher starch content (28.5 vs 14.2%) in the first 45 days of life did not affect rumen fermentation, rumen microbiota and the most important blood metabolites, apart from a few exceptions. The production parameters of calves, including total feed intake and body weight gain, were similar in the experiment. |

Chickens were kept in two pens on wooden shavings (2 × 96) from one day of age. On day 23 of age, one group was moved to a pasture and kept in a portable floorless pen with an area of 0.11 m2 per chicken. The field portion of the experiment was conducted from the 1st till the 18th of June 2013. A control group was kept in the original pen until the end of the experiment at 6 weeks of age. Both groups were fed the same pelleted feed ad libitum. For the pasture-reared group, the moveable shelter was moved twice daily around the pasture with a predominance of grass species. The herbage intake of chickens was indirectly assessed by compressed sward height measurement after each cage movement, which employed a rising plate-meter. In the pasture, chickens preferred grass over clover. There was no significant effect of grazing observed on chicken body weight. The mortality of chickens in the grazing group was lower than that in the control group. Pasture treatment improved meat flavour by 9% (P = 0.014), produced breast meat with significantly higher (P = 0.009) redness, and almost doubled the concentration of α-tocopherol (P < 0.001). There were no significant effects of grazing on the dry matter, fat, cholesterol or pH of the meat. The production of TBARS in the breast meat of the pasture group after storage at 4°C for 5 days was lower (P = 0.013) than that in the breast meat of the control chickens. Although the differences between K, Ca, P, Mg, and N concentrations in pasture and soil before and after grazing reached 20%, these differences were not significant. |

Sci., 2015, 60(9):391-399 | DOI: 10.17221/8457-CJAS Polymorphic SNPs were identified using BovineSNP50 BeadChip in three groups of cervids: farmed Red deer (n = 3), and free range Red deer (n = 5) and Fallow deer (n = 2). From the total of 54 609 SNPs, 53.85% could be genotyped. Out of 28 502 successfully genotyped autosomal SNPs only 5.3% were polymorphic. The average minor allele frequency within cervids was 0.23 (number of polymorphic SNPs ranged from 467 to 686). Results of the molecular variance analysis showed that 67.38% of variation occurred within individuals and the rest was explained by a species difference (FST = 0.32). The value of FIT (0.33) indicated a higher proportion of homozygote genotypes in the analyzed dataset. Pairwise FST values showed very clearly the genetic differentiation between Red and Fallow deer which ranged from 0.06 (farmed and free range deer) to 0.74 (farmed Red and Fallow deer). A similar result was found for Nei's genetic distances that ranged from 0.01 (among Fallow deer) to 0.79 (among farmed Red and Fallow deer). The genetic differentiation of the analyzed cervid species was evaluated also by the principal component analysis with the involvement of 6 other species from the family Cervidae, which showed a division of the Cervidae cluster into 7 subpopulations. The panels of SNPs primarily produced for a model species are becoming the marker of choice for the application in other species, but the best methods of their discovery, validation, and genotyping in non-model species need further investigations. |
