Sci., 2023, 68(12):486-496 | DOI: 10.17221/89/2023-CJAS The use of genomic data makes it possible to examine genetic variability and calculate the genetic parameters of the population in an efficient and precise way. The aim of this research was to analyse linkage disequilibrium (LD), contemporary effective population size (Nec), haplotype block structure, minor allele frequency (MAF), observed (HO) and expected heterozygosity (HE), calculate the genomic relationship matrix and perform a principal component analysis (PCA) in the Serbian Holstein-Friesian cattle population using SNP data from the GGP Bovine 100K chip. After quality control (QC), 83 208 SNPs and 1 575 cows were retained for further analysis. LD on autosomes had an average value of ≥ 0.2 up to a distance of 50–60 kb (r2 = 0.211), while on BTX r2 ≥ 0.2 was represented at distances of 80–90 kb (r2 = 0.211). LD differed between chromosomes. The average HO for autosomes and X chromosome SNPs was 0.412 and 0.422, respectively. 81.30% of SNPs that passed QC had MAF > 0.2. The total number of haplotype blocks in the studied population was 15 642. On average, blocks contained 2.932 SNPs. The average block length was 32.657 kb and ranged from a minimum of 0.019 kb (BTA21 and BTA26) to a maximum of 999.562 kb (BTX). The estimated value of Nec in the this cattle population was 142. The results of PCA showed a significant variability of genotypes in the population, but there was no clear stratification of the population. The obtained results will serve as a basis for future genomic analyses such as the detection of QTLs for important economic traits and the implementation of genomic selection. |
Effect of catalase on the growth performance, antioxidation, and microbial metabolism of weaned rabbitsOriginal PaperNing Liu, Yien Lin, Shuaibao Wang, Jianping WangCzech J. Anim. Sci., 2023, 68(12):508-515 | DOI: 10.17221/133/2023-CJAS The present study aimed to investigate the effect of catalase (CAT) on the growth performance, antioxidation, and microbial metabolism of weaned rabbits. Dietary treatments consisted of control and CAT supplementation at 100 (T1), 150 (T2), and 200 IU/kg of diet (T3). A total of 240 weaned rabbits were randomly assigned to 4 groups with 6 replicates of 10 rabbits each. The feeding trial lasted for 28 days. Results showed that T2 and T3 increased (P < 0.05) body weight gain and gain/feed ratio, and decreased (P < 0.05) diarrhoea rate, compared to the control. Also, serum antioxidative parameters and oxidative stress products were beneficially regulated (P < 0.05) by the dietary CAT administration. Faecal microbiota including Bacteroidetes, Prevotella, and Bifidobacterium in T2 or T3 was increased (P < 0.05). Dietary CAT with changed microbiota further influenced the metabolites from carbohydrates and proteins, evidenced by increased lactic acid, acetate, branched-chain fatty acids, and short-chain fatty acids, and decreased valerate, isovalerate, methylamine, tryptamine, putrescine, cadaverine, spermidine, and total amines. It is concluded that CAT can be used to improve growth performance by beneficially regulating the antioxidation, microbiota, and metabolites in weaned rabbits. |

Sci., 2024, 69(8):323-330 | DOI: 10.17221/91/2023-CJAS Indigenous chicken breeds from Fujian Province, China, exhibit unique and distinctive phenotypic characteristics, including morphology, behaviour, integumentary pigmentation, feather colouration, and production performance. However, the underlying genetic basis governing these specific traits remains unclear. This study aimed to elucidate the population structure of six Fujian local chicken breeds [Bairong chickens (BRCs), Jinhu chickens (JHCs), Minqing chickens (MQCs), Dehua chickens (DHCs), Xiangdong chickens (XDCs), and Hetian chickens (HTCs)] in terms of their genetic resources within the province. The whole-genome resequencing revealed 8 678 612 single nucleotide polymorphisms (SNPs) in a cohort of 36 chickens. The results revealed a partial mismatch between the clustering patterns and geographic distributions of the six populations, with BRC displaying a noticeable genetic divergence from other chickens. Moreover, the genetic distance between XDC and HTC in Longyan County was relatively close and converged with JHC, MQC, and DHC into a subgroup. These findings provide a comprehensive understanding of the intrinsic characteristics and utilisation prospects of chicken germplasm resources in Fujian Province. |

Conditions for cold stress development in dairy cattle kept in free stall barn during severe frostsOriginal PaperS. Angrecka, P. HerbutCzech J. Anim. Sci., 2015, 60(2):81-87 | DOI: 10.17221/7978-CJAS The impact of low temperatures combined with higher velocity of ventilated air in winter during severe frost in a free stall barn on the development of cold stress in Holstein-Friesian breed dairy cattle was studied. The study included measurements of outside and inside air parameters and cows' milk yield. Cold stress was evaluated in three technological groups (TG) using the WCT (Wind Chill Temperature) index. During the research, significant temperature and wind velocity differences (by up to 5°C and 0.5 m/s) were recorded in three zones of the barn occupied by the individual TG. All this resulted in different values of WCT. During most severe frosts lasting for 9 days, the average air temperature in the barn was -8.9°C (the lowest value was -17.3°C). The calculated average operative temperature for cows from all TG was as follows: -7.0°C for TG1, -11.1°C for TG2, and -12.3°C for TG3. It was also observed that animals from TG2 suffered from mild cold stress, which resulted in milk yield reduction by approximately 2 kg. It was concluded that there is a strong correlation (r = 0.72-0.89 with P < 0.05) between milk production and the WCT index. The measurements of most important microclimate parameters in the barn were conducted during winter seasons over the period of 2 years. They led to the conclusion that cattle kept in free stall barns are not vulnerable to the combination of low temperature and increased air movements. Research results of the present as well as of other authors were the basis for developing a table determining operative temperature for cows depending on the temperature of the environment and air movement velocity. Yet, the calculations and analysis of results show that there is a need to improve the applied calculation formula for operative temperature during cold weather. |

This research aimed to evaluate the effect of protease on nutrient digestibility, amino acids, and metabolism of AOM energy for broilers. Four hundred and eighty broiler chickens were distributed in a completely randomized design (4 × 2 factorial scheme), eight treatments, six replicates containing 10 birds/replicate. Treatments consisted of poultry viscera meal, swine viscera meal (SVM), bovine meat and bone meal, and basal diet; with and without protease addition. Two tests were performed. In the first test, the total excreta collection method was used with birds at 13 to 20 days of age and 25% of the reference feed was replaced by AOM. In the second test, a protein-free diet was administered to birds at 21 to 24 days of age and AOM replaced 25% of the starch. The inclusion of protease increased the apparent metabolizable energy corrected for nitrogen balance of SVM by 15.99% and the apparent metabolizable crude energy by 5.7%, and it also raised the coefficient of true ileal digestibility of the amino acids in the AOMs by 5.67% on average. The inclusion of protease improved the apparent metabolizable crude energy of AOMs, apparent metabolizable dry matter of bovine meat and bone meal, coefficient of true ileal digestibility of essential amino acids, and apparent metabolizable energy corrected for nitrogen balance of SVM. Dietary supplementation of protease may be a potential strategy to improve the digestibility of amino acids for broilers, a possibility of using animal-origin meals as a protein source of diets. |

Sci., 2021, 66(9):368-380 | DOI: 10.17221/184/2020-CJAS This study assessed the effect of the combination of microencapsulated sodium butyrate (SB) and phytase (PH) on the performance, intestinal integrity, and bone development of broilers. The experiment comprised 736 chicks distributed in a completely randomised design in a 2 × 2 factorial scheme (with and without the inclusion of SB and with the inclusion of PH at the recommended dose and superdosing) totalling four treatments and eight repetitions of 23 birds each. SB was added at 0.750 kg/tonne and PH was included at 750 phytase units (FTU)/kg and 1 500 FTU/kg of diet. The live weight (LW), feed intake (FI) and feed conversion (FC) were evaluated weekly. Bone measurements of the tibia and femur, densitometry, length, the Seedor index, and diameter were performed at seven and 28 days. In the same ages, fragments of the jejunum and ileum segments were collected and subjected to a morphometry analysis. At seven days of age, the birds supplemented with the phytase superdosing showed a better FI and LW, and the microencapsulated SB in the diet showed a better LW and FC. In the period from one to 28 days, the treatment supplemented with SB provided a higher FI and LW. The SB supplementation resulted in greater bone measurements at seven and 28 days. The use of the superdosing phytase increased the villus length and width of the jejunum at seven days and at 28 days. There was an increase in the villus length, villus-to-crypt ratio, absorption area, and muscular layer of the jejunum and a decrease in the crypt width of the jejunum. The SB supplementation increased the ileum crypt width at seven days and there was no effect on any intestinal segment at 28 days. The supplementation of SB or PH 1 500 FTU/kg in the broiler diets' growth promoter or antibiotic-free resulted in a better performance and bone measurements, while the association of the additional PH 1 500 FTU/kg and SB supplementation resulted in the greater depth and width of the jejunum crypt and villus : ileum crypt at seven days and greater villus length and ileum absorption area at 28 days. The association of SB and PH may be a strategy to improve the performance and bone quality and intestinal integrity in broilers. |

Serum concentrations of proline, alanine, tyrosine and aromatic amino acids were significantly higher in males than in females (all P = 0.05). Taurine and glycine concentrations in skeletal muscles were significantly higher in males than in females (P < 0.01). Muscle concentrations of cysteic acid, taurine, aspartate, threonine, serine, glycine, alanine, citruline, valine, leucine, gamma-amino-butyrate, ethanoloamine, lysine and histidine were significantly higher than in serum in both sexes (P < 0.05). In females, the concentrations of glutamate, glutamine, cystathionine, isoleucine, tyrosine, phenylalanine, arginine and amino-adipic acid were significantly higher in muscles than in serum (P < 0.05). Tryptophan concentration was significantly higher in serum from males than in muscles (P = 0.01). The concentration of branched-chain amino acids in skeletal muscles was approximately two times higher than in serum in both groups of foxes (P ≤ 0.01). Similar differences were obtained for aromatic amino acids in females (P = 0.04). The elaborated experimental model may serve for further studies focused on amino acid metabolism regulation in Canide and other monogastric mammals, especially with the use of environmental, dietary, pharmacological and toxicological factors. The elaborated experimental model may be an attractive alternative to replace some experiments on dogs. |

Sci., 2022, 67(12):475-482 | DOI: 10.17221/149/2022-CJAS Dairy farming is deeply affected by climate change, especially by rising temperatures and heat waves, poorer availability of quality food and water, and the spread of new diseases and pests outside their original ecological niche. Their impact can be mitigated not only by changes in technologies, management and treatment, but also by breeding and selection of more resilient cows. General resilience encompasses the animal's capacity to cope with environmental, social and disease challenges. It is described as the capacity of the animal to be minimally affected by a disturbance or to rapidly return to the physiological, behavioural, cognitive, health, affective and production states that pertained before exposure to a disturbance. As disturbances can be of different natures, general resilience is a composite trait consisting of different resilience types according to the nature of the disturbance. Resilience can be quantified through time series data that capture fluctuations in the daily performance. Recent studies have worked with deviations in the daily milk yield and daily live weight from optimal performance or have focused on the assessment of the daily activity in terms of the daily step count. To observe the duration and magnitude of the response to perturbance, two indicators were suggested: the autocorrelation (rauto) and the natural logarithm of deviations (LnVar). Based on the daily milk yield deviations, both indicators have shown sufficient genetic variabilities with the estimated heritability ~0.1 for rauto and ~0.2 for LnVar. Low values of both indicators were genetically related to better udder health, better hoof health, better longevity, better fertility, higher body condition score, less ketosis but also lower milk yield level. The selection for improved resilience could benefit from the use of genomic information as several genes and biological pathways associated with disease resilience and resilience to heat stress have already been identified. The presented results suggest that the integration of resilience into the cattle breeding programmes would improve the capacity of the dairy industry to cope with global climate change. |
Effects of dietary addition of wormwood on growth performance, blood characteristics and meat quality in growing-fattening pigsOriginal PaperGyo Moon Chu, Byung Ki ParkCzech J. Anim. Sci., 2022, 67(12):503-514 | DOI: 10.17221/110/2022-CJAS This study investigated the effects of the dietary addition of wormwood (Artemisia montana Pampan) on growth performance, blood characteristics, carcass traits, and meat quality in growing-fattening pigs. One hundred eighty crossed pigs (Landrace × Yorkshire × Duroc), weighing approximately 70 kg, were divided into four groups of 15 head (eight barrows and seven gilts) per pen, and the experiment was replicated thrice. The basal diet (C) was supplemented with 0.5% (T1), 1.0% (T2), and 1.5% (T3) of dried powdered wormwood, and the pigs were fed an experimental diet for six weeks. The average daily weight gain and feed efficiency were significantly higher (P < 0.05) in treatments than in C. On the contrary, the total cholesterol concentration was significantly lower (P < 0.05) in treatments than in C. Additionally, the high-density lipoprotein cholesterol concentration was significantly higher (P < 0.05), and the carcass grade was better (P < 0.05) in the treatments than in C. The ether extract content of the longissimus dorsi (LD) muscle was lower (P < 0.05) in T2 and T3 than that in C. In contrast, the unsaturated fatty acid (USFA) composition in LD muscle was higher (P < 0.05) in the treatments than in C. Additionally, the marbling and flavour of cooked meat were better (P < 0.05) in the treatments than in C. The dietary addition of wormwood increased (P < 0.05) lightness and yellowness of the surface meat colour and decreased (P < 0.05) juiciness of the LD sensory evaluation. Therefore, the dietary addition of wormwood improved growth performance, feed efficiency, carcass grade, USFA composition, and oxidation of protein and lipids in the LD muscle of growing-fattening pigs. |

After being withered a little, the fodder plant was fragmented into 2–3 cm long pieces. 0, 2 and 4% molasses and 0, 0.5 and 1% urea were added. The treatment groups were UM1 (control), UM2, UM3, UM4, UM5, UM6, UM7, UM8 and UM9 supplemented with 0, 0, 0, 1, 1, 1, 1.5, 1.5, 1.5% urea and 0, 2, 4, 0, 2, 4, 0, 2, 4% molasses respectively, and a total of 54 silage samples, 9 × 6 parallel, were prepared, vacuumed in ready-made silage bags, and stored at 25 ± 2 °C for 60 days. After the silages were opened, pH, nutrient contents, Fleig scores (FS), in vitro digestibility parameters, energy contents, feed value and feed quality were determined, then the data were subjected to analysis of variance. The effects of urea and molasses additives added to the mixed forage plant silage on all parameters were found to be significant (P < 0.05, P < 0.01). The crude protein (CP) increased its application by UM7. Cell wall component (insoluble fibre in acid detergent (ADF) and neutral detergent insoluble fibre NDF) reduced its application by UM9. All these applications increased the FS. The addition of molasses increased the in vitro gas production, while the addition of urea decreased the production. True organic matter digestibility increased with the addition of molasses. As a result, it was concluded that UM9 application in silages would be appropriate for increasing feed value and feed quality, and the UM3 application in silage would be appropriate for improving true organic matter digestibility (TOMD). |

Sci., 2023, 68(8):333-345 | DOI: 10.17221/2/2023-CJAS Seasonal heat stress and metabolic disorders during mid-lactation are the main factors limiting production in Holstein cows, and several proteins and molecules involved in metabolic pathways are altered in response to stress. We investigated the effects of heat stress on the milk yield, milk composition, serum oxidative status, and metabolites in Holstein cows during mid-lactation to identify biomarkers associated with heat stress in serum and milk. Holstein cows with similar body condition scores (3.0 ± 0.25), parity (2.5 ± 0.5), and lactation days (115 ± 5 days) were selected in August (heat stress, HS, n = 20) 2017 and March 2018 (non-heat stress, NHS, n = 20). Milk yield was recorded daily and serum was collected on days 1, 31, and 61. Serum and milk metabolites were analysed by gas chromatography-mass spectrometry on day 1. The results showed a significantly lower average daily milk yield in the HS group than in the NHS group (P < 0.05). The milk compositions of fat (%), lactoprotein, lactose yield, and milk solid-not-fat in the HS group were significantly lower than in the NHS group on days 1, 31, and 61 (P < 0.05). The levels of malondialdehyde were higher, whereas those of superoxide dismutase and glutathione peroxidase were lower in the serum of the HS group (P < 0.05) than that of the NHS group. The serum concentrations of d-glucose, 9,12-octadecadienoic acid, and d-lactose were significantly higher in the NHS group than in the HS group (P < 0.05). The concentrations of lactic acid and milk urea nitrogen in the NHS group were lower than those in the HS group (P < 0.05). The present data suggest that metabolic biomarkers are closely associated with heat stress in the serum and milk, which provides a basis for evaluating indicators of heat stress occurrence in mid-lactation cows. |

Sci., 2023, 68(9):383-390 | DOI: 10.17221/4/2023-CJAS This study was conducted to determine the individual types of crystallisation structures of cervical mucus (arborisation phenomenon) in sheep and changes in the types of these structures during oestrus. A total of 80 ewes aged between two and eight years were included in the study. Oestrus synchronisation was performed using intravaginal sponges, and samples of cervical mucus were collected 55–57 h after the removal of intravaginal sponges and subsequently evaluated. The crystallisation structures of the cervical mucus of sheep exhibit the same sequence during oestrus as in cattle. The most frequently occurring types of crystallisation in the observed sheep included twig-shaped (33.33%), twig-shaped-clubmosses (24.00%) and clubmosses (14.67%). The least common type was fern frond crystallisation (1.33%). A statistically significant difference was found between the representation of individual types of crystallisation (P < 0.01). As with cattle, it is possible to use the evaluation of the crystallisation of cervical mucus to determine the optimum time for insemination, especially in groups of animals with previous synchronisation/induction of oestrus. Establishing the correct time for insemination or natural mating with a ram makes it possible to optimise breeding management and improve pregnancy rates, which are crucial in breeding these seasonally polyoestrous animals. Based on the results, it can be stated that insemination should take place 57+ h after the removal of intravaginal sponges and the application of hormonal treatments for oestrus induction and synchronisation (equine chorionic gonadotropin). |

Sci., 2023, 68(9):372-382 | DOI: 10.17221/193/2022-CJAS The aims of this study were to examine bacterial communities in relation to the rumen digestion of rice straw and to understand how concentrate supplements affect gut bacteria involving the digestion of a rice straw-based diet. The substrates were rice straw (RS) alone (experiment 1) and RS with 25% concentrates (barley and kidney beans) (experiment 2). The genomic DNA was collected to determine bacterial diversity by conducting denaturing gradient gel electrophoresis (DGGE). V6–V8 region group-specific (Clostridium and Bacteroides) primers were employed in the analyses. The DGGE band pattern was subjected to cluster analysis to demonstrate the similarity and difference between dietary treatments and solid-liquid fractions. Fibre digestibility, gas production, and volatile fatty acid (VFA) concentration were increased with incubation time. The differences between solid and liquid fractions were great in total bacteria, Clostridium, and Bacteroides communities. Clostridium and Bacteroides communities appeared unaffected by incubation time, whereas great differences existed between solid and liquid fractions throughout incubations (experiment 1). Barley and kidney bean supplements increased gas production and lowered rumen fluid pH, whereas changes in VFA concentration were significant only for kidney bean supplement. The Clostridium, and Bacteroides groups were affected by barley and kidney beans in the liquid fraction. However, the solid fraction was stable (experiment 2). These results indicate a rigid and stable community structure of Clostridium and Bacteroides groups involved in the digestion of rice straw-based diets in the rumen. Distinctive differences between solid and liquid fractions were described; hence, separate analyses of two fractions may greatly help understand the relationship between fermentation patterns and microbial communities in the rumen. |
Polymorphism of the PGLYRP1 gene, the value of selected performance and functional traits, and causes of culling in Holstein-Friesian red-white cowsOriginal PaperPiotr Sablik, Andrzej Dybus, Ewa Janu¶, B³a¿ej Nowak, Wojciech Kruszyñski, Anna MuchaCzech J. Sci., 2023, 68(10):399-406 | DOI: 10.17221/194/2022-CJAS This research paper addresses the hypothesis that the peptidoglycan recognition protein 1 (PGLYRP1) gene polymorphism (Tyr76His; dbSNP ID: ss104796364) has an influence on some performance traits and causes of culling in Polish Holstein-Friesian red-white cows. The study involved 134 cows kept on a farm in the south-west of Poland. PGLYRP1 genotypes TT, CT, and CC were detected. It was shown that compared with cows with genotypes CT and TT, the individuals with genotype CC were characterised by higher lifetime yields and higher amounts of lactation milk, fat, and protein. A beneficial effect of genotype CC, compared with genotype TT, was also noted in the case of the lifespan and, consequently, the length of the productive life and the average number of lactations. Diseases of the musculoskeletal system (genotypes CC and TT) and disorders of the reproductive system (genotype CT) were the most common causes of culling. An essential practical observation was the potentially higher susceptibility of cows with genotype CC to mastitis, which resulted in the necessity to cull over one-fifth of the animals in this group. Simultaneously, no cows in this group were culled due to low performance or metabolic, gastrointestinal, and respiratory diseases. Therefore, the PGLYRP1 gene seems to be a promising potential herd health marker; however, to consider it the main gene, it is necessary to extend the investigations to include more individuals and other breeds of dairy cattle. |
Growth, carcass and meat quality in Zwartbles lambs slaughtered at different live weightsOriginal PaperTomá¹ Jano¹, Jan Kuchtík, Eli¹ka Draèková, Martin Ho¹ek, Tomá¹ Kopec, Radek FilipèíkCzech J. Sci., 2023, 68(10):414-422 | DOI: 10.17221/99/2023-CJAS The aim of the study was to evaluate the effect of different live weights at slaughter (LWS) of Zwartbles lambs on their daily gain (DG), carcass traits (CT) and chemical and physical characteristics of the quadriceps femoris muscle (QFM). A total of four weight groups of lambs were evaluated: group A, LWS = up to 35 kg; group B, LWS from 35.1 to 40 kg; group C, LWS from 40.1 to 45 kg and group D, LWS from 45.1 to 50 kg. The LWS had a significant (P < 0.05) effect on DG and most CTs, when average DG and most weights of individual CTs increased with increasing LWS. The evaluation of the influence of LWS on the chemical and physical characteristics of QFM primarily shows that this factor had a significant (P < 0.05) effect only on the content of intramuscular fat (IMF) and redness index (RI), when in both cases the values of these traits increased (IMF: from 0.57 to 1.21%; RI: from 8.53 to 9.76) with increasing LWS. In conclusion, it can be stated that most of the monitored traits in all weight groups of Zwartbles lambs were comparable with their levels in specialized meat breeds of sheep. |

Nevertheless, dairy farmers are challenged increasingly by the consequences of global warming. Economic risks like feed supply and volatile commodity prices need to be balanced, also taking into account the increasing environmental awareness of end-customers. Focusing just on emissions, dairy production is contributing an essential part of the total carbon footprint emitted by the agricultural sector. Sci., 2020, 65(11):411-423 | DOI: 10.17221/189/2020-CJAS Modern single space feeding systems for fattening pigs allow the detailed assessment of an individual animal's feeding behaviour. In an experiment involving 96 fattening pigs, the influence of deoxynivalenol (DON) contaminated feed (> 4 500 µg/kg DON) on the zootechnical performance and feeding behaviour was compared with a feed with low DON concentration (< 900 µg/kg DON), this served as the control group. Additionally, in separate treatments, two commercial mycotoxin binders were added to the DON-contaminated feeds to assess if an expected DON effect could be attenuated. The high DON content significantly (P < 0.03) reduced daily feed intake (500-600 g/day). The DON group showed 240 g less daily gain compared to the control with 728 g/day. Both mycotoxin binders were seen to additionally depress weight gain by approximately 65 g/day (P < 0.01). The treatment did not affect the individual feeding behaviour as assessed by daily visits to the feeder, feed intake per visit and the highest feed intake per visit per day. These were influenced only by the pig and its pen, indicating that the animals developed a distinct behaviour within their respective groups. Behaviour analyses of persistency and day-to-day variation showed that the persistency was reduced and variation was increased when DON without or with binder was present. The DON contents therefore seemed to lead to a more erratic and less consistent behaviour that remained dependent on the animal group. |
