Temperature regulates fatty acid desaturase and elongase at the transcriptional level and modulates the fatty acid profile in the early stage of the common carp (Cyprinus carpio)Original PaperHong-Tao Ren, Shi-Yang Gao, Yong Huang, Xiao-Chan GaoCzech J. Sci., 2023, 68(7):313-321 | DOI: 10.17221/22/2023-CJAS This study explored the regulatory effect of temperature stress on fatty acid biosynthesis in the early stage of common carp (Cyprinus carpio) based on Δ6-fatty acids desaturase (Δ6FAD) and elongase-5 (ELOVL5) gene expression and fatty acid composition. One-day-old carp larvae were selected, and after seven days of acclimatisation at 25 °C, the larvae were subjected to temperature stress for 96 h in water at 32 °C or 10 °C. In the post-larval stage, 30-day-old carp juveniles were selected and, after seven days of acclimatisation at 25 °C, were subjected to temperature stress for seven days in water at 32 °C or 10 °C. The results showed that common carp larvae could rapidly and highly express Δ6FAD and ELOVL5 genes within 48 h at high temperature (32 °C) compared with the 0 h group (P < 0.05), while gene expression began to gradually increase after 48 h at low temperature (10 °C). There was a significant improvement in C22:6n-3 and C20:5n-3 after 96 h at low temperature compared with the 0 h group (P < 0.05). In common carp juveniles, the Δ6FAD gene in the intestine, brain and liver was sensitive to low temperature, but the ELOVL5 gene in the intestine, brain and liver of common carp was sensitive to high temperature. The low temperature increased the amount of highly unsaturated fatty acids (HUFA) in the common carp juveniles. The results indicated that temperature could regulate the expression of the Δ6FAD and ELOVL5 genes for HUFA production, as well as for participation in the biosynthesis of fatty acids in the body during the early development of common carp. The results of this study help clarify the regulatory effects of temperature on fatty acid biosynthesis during the early development of common carp. |

This study was conducted to assess the genetic diversity as well as the phylogenetic relationships of Vietnamese indigenous chickens (Gallus gallus) to support the conservation of their genetic resources for sustainable rural farming. In this study, the genetic diversity and the phylogenetic relationships of 10 Vietnamese indigenous chicken breeds were analyzed using complete sequences of mitochondrial DNA (mtDNA) displacement-loop (D-loop) region. The average nucleotide and haplotype diversities of Vietnamese chickens were 0.006 3 ± 0.000 22 and 0.918 ± 0.010, respectively. A total of 39 polymorphic sites and 29 haplotypes were identified. The maximum likelihood tree classified these haplotypes into seven haplogroups (A, B, C, D, E, G and V), with haplogroups A and B being the two predominant maternal lineages of Vietnamese indigenous chickens, while haplogroups C, D, E, G and V were found in the remaining chickens. Several haplotypes from different haplogroups were shared among some chicken breeds. These results suggested that Vietnamese indigenous chicken breeds have multiple maternal origins, mainly from Chinese, Southeast Asian and Indian chickens, and that these breeds share common maternal lineages. The high level of genetic diversity in Vietnamese chickens demonstrates significance of conservation for future use. |

Sci., 2023, 68(7):296-305 | DOI: 10.17221/148/2022-CJAS This study is to explore the effects of natural tannin (NBT) on intestinal injury in piglets challenged with enterotoxigenic Escherichia coli (ETEC). Thirty-two 7-day-old piglets were divided into four groups: I) control group, piglets were fed a basic diet without challenge; II) ETEC group, piglets were fed a basic diet and challenged with enterotoxigenic E. coli; III) NBT + ETEC group, piglets were fed a basic diet with supplementation of 2 g/kg NBT and challenged with enterotoxigenic E. coli; and IV) ZnO + ETEC group, piglets were fed a basic diet with supplementation of 3 g/kg ZnO and challenged with enterotoxigenic E. coli. Results showed that diarrhoea rate was significantly increased in ETEC group, whereas it was decreased after NBT supplementation (P < 0.05); NBT supplementation significantly increased the average daily gain of ETEC-infected piglets. ETEC group showed damaged intestinal morphology (as indicated by decreased villus height, surface area and increased crypt depth) and barrier function (as indicated by decreased d-xylose content and increased diamine oxidase activity), induced oxidative stress (as indicated by increased myeloperoxidase activity) while supplementation of NBT had an obvious impact on the recovery of intestinal function and alleviated the oxidative damage. Further analysis showed that NBT could decrease the expression of genes related to intestinal injury (matrix metalloproteinase-3), and specifically upregulate the expression of oxidative stress-related genes (nuclear factor erythroid 2-related factor 2 and hypoxia inducible factor-1). Overall, dietary supplementation of NBT relieved intestinal injury and oxidative stress in piglets challenged with ETEC. NBT could be an alternative to ZnO as a feed additive in piglet diet. |

Milk samples were collected from the group of 38 non-dairying sheep selected from the basic 120-head flock. Ewes were monitored during four control days (from 27th April, the average 42nd day of lactation to 4th August, the average 142nd day of lactation) to cover the whole lactation period. Ewes were investigated for their milk production and for milk quantity parameter. Milk quality indicators included percentage estimation of milk components (fat, protein, casein, lactose, dry matter), somatic cell count (103 cells/ml), vitamin A and E content (mg/kg), and K, Mg, Ca, Zn, Cu (mg/kg) content. Milk production ranged from 1 017.37 g (early stage of lactation) to 416.87 g (late stage of lactation period). The milk contained high fat (6.06–8.44%) and protein (4.68–5.68%) percentages, low somatic cell count and favourable distribution of minerals and vitamins. In general, results of this study indicate a possibility for low-productive traditional grazing system with extensive sheep breeds not only for cultivating the area but also for the purpose of high nutritional food with beneficial aspect on human health. |

Anim. Sci., 2023, 68(11):443-450 | DOI: 10.17221/107/2023-CJAS Lipid-rich bovine serum albumin (LR-BSA) has been reported to increase the survival rate of blastocysts after freezing and thawing. However studies on the early development of in vitro fertilised (IVF) embryos are still insufficient in cattle. This study investigated the blastocyst productivity of in vitro culture (IVC) medium (synthetic oviductal fluid medium with amino acids, mSOFaa) supplemented with 5% fetal bovine serum (FBS, Control) or LR-BSA (1, 2.5, 5 mg/ml dosages, respectively) and the qualitative characteristics of produced blastocysts. Cleavage rates on day 2 were similar between all groups. Significant differences were obtained in the blastocyst rate in the 2.5LR-BSA group compared with the control (23.5% vs 35.7%, P < 0.05). Hatching rates were significantly higher in all treatment groups than in control (P < 0.05). The cell number in blastocysts did not differ among groups, but the apoptotic index in the 5.0LR-BSA group was higher than the control (P < 0.05). The expression of embryo quality-related markers was affected. Octamer-binding transcription factor 4 (Oct4), placenta associated 8 (Plac8), and sex-determining region Y-box 2 (Sox2) genes expression in the 2.5LR-BSA group were significantly up-regulated compared to the control (P < 0.05). Cytoplasmic lipid contents and the lipid droplet formation-related gene, sterol regulatory element binding transcription factor 1 (Srebf1), were significantly increased in the 5.0LR-BSA group (P < 0.05). Therefore, the LR-BSA supplementation (2.5 mg/ml) to mSOFaa medium could contribute to the early development of bovine IVF embryos and the qualitative improvement of the resulting blastocysts. |

Sci., 2023, 68(9):383-390 | DOI: 10.17221/4/2023-CJAS This study was conducted to determine the individual types of crystallisation structures of cervical mucus (arborisation phenomenon) in sheep and changes in the types of these structures during oestrus. A total of 80 ewes aged between two and eight years were included in the study. Oestrus synchronisation was performed using intravaginal sponges, and samples of cervical mucus were collected 55–57 h after the removal of intravaginal sponges and subsequently evaluated. The crystallisation structures of the cervical mucus of sheep exhibit the same sequence during oestrus as in cattle. The most frequently occurring types of crystallisation in the observed sheep included twig-shaped (33.33%), twig-shaped-clubmosses (24.00%) and clubmosses (14.67%). The least common type was fern frond crystallisation (1.33%). A statistically significant difference was found between the representation of individual types of crystallisation (P < 0.01). As with cattle, it is possible to use the evaluation of the crystallisation of cervical mucus to determine the optimum time for insemination, especially in groups of animals with previous synchronisation/induction of oestrus. Establishing the correct time for insemination or natural mating with a ram makes it possible to optimise breeding management and improve pregnancy rates, which are crucial in breeding these seasonally polyoestrous animals. Based on the results, it can be stated that insemination should take place 57+ h after the removal of intravaginal sponges and the application of hormonal treatments for oestrus induction and synchronisation (equine chorionic gonadotropin). |

Sci., 2023, 68(10):407-413 | DOI: 10.17221/39/2023-CJAS The aim of this study was to estimate the effects of crossbreeding on milk production and milk components in crossbred Tsigai, Lacaune, and Improved Wallachian sheep used to create a synthetic Slovak dairy sheep population. The local breeds Tsigai and Improved Wallachian were crossed with the high-yielding breed Lacaune to increase milk yield. The effects of crossbreeding were evaluated based on breed and heterosis effects. The database used consisted of 23 018 lactations from 15 888 ewes. The effects of crossbreeding on milk production and fat and protein content were estimated using the restricted maximum likelihood method in REMLF90. Fixed effects on milk yield were also studied. In ewes with a 100% heterosis effect (ewes from purebred parents of different breeds), the heterosis effect increased milk production by 11.642 kg, milk fat production by 0.772 kg, and protein production by 0.565 kg per milking period. The effect of the Tsigai breed on milk production was –21.98 kg compared to Lacaune, while for the Improved Wallachian breed, the value was –38.58 kg. The Tsigai direct breed effect reduced fat production by 1.56 kg and protein production by 1.34 kg. The Improved Wallachian direct breed effect was –2.71 kg for fat production and –2.27 kg for protein production. |

A total of 240 one-day-old Arbor Acres broiler chickens were randomly allocated to four treatments, with six replicates per treatment and 10 broilers per replicate. Chickens were fed corn-soybean meal basal diets supplemented with 0, 4, 8, and 12 IU/g of protease. Compared with 0 IU/g of protease supplementation, 12 IU/g of protease increased the average and minimum time of feeding and drinking behaviour (P < 0.05); increased the average, maximum, and minimum time of preening behaviour (P < 0.05); and decreased feeding, walking, and stretching frequency (P < 0.05). pH24h of breast and thigh muscles had improved (P < 0.05). The shear force of the breast muscle and the cooking loss of the thigh muscle had decreased (P < 0.05). Compared with 0 IU/g of protease, 4 IU/g of protease increased the average and minimum time of lying behaviour (P < 0.05) and decreased stretching, scratching, and aggressive frequency (P < 0.05). The shear force of the thigh muscle and spleen index had enhanced (P < 0.05). Protease can be used as an additive in a broiler diet for production. |

Sci., 2023, 68(10):399-406 | DOI: 10.17221/194/2022-CJAS This research paper addresses the hypothesis that the peptidoglycan recognition protein 1 (PGLYRP1) gene polymorphism (Tyr76His; dbSNP ID: ss104796364) has an influence on some performance traits and causes of culling in Polish Holstein-Friesian red-white cows. The study involved 134 cows kept on a farm in the south-west of Poland. PGLYRP1 genotypes TT, CT, and CC were detected. It was shown that compared with cows with genotypes CT and TT, the individuals with genotype CC were characterised by higher lifetime yields and higher amounts of lactation milk, fat, and protein. A beneficial effect of genotype CC, compared with genotype TT, was also noted in the case of the lifespan and, consequently, the length of the productive life and the average number of lactations. Diseases of the musculoskeletal system (genotypes CC and TT) and disorders of the reproductive system (genotype CT) were the most common causes of culling. An essential practical observation was the potentially higher susceptibility of cows with genotype CC to mastitis, which resulted in the necessity to cull over one-fifth of the animals in this group. Simultaneously, no cows in this group were culled due to low performance or metabolic, gastrointestinal, and respiratory diseases. Therefore, the PGLYRP1 gene seems to be a promising potential herd health marker; however, to consider it the main gene, it is necessary to extend the investigations to include more individuals and other breeds of dairy cattle. |

Sci., 2023, 68(9):372-382 | DOI: 10.17221/193/2022-CJAS The aims of this study were to examine bacterial communities in relation to the rumen digestion of rice straw and to understand how concentrate supplements affect gut bacteria involving the digestion of a rice straw-based diet. The substrates were rice straw (RS) alone (experiment 1) and RS with 25% concentrates (barley and kidney beans) (experiment 2). The genomic DNA was collected to determine bacterial diversity by conducting denaturing gradient gel electrophoresis (DGGE). V6–V8 region group-specific (Clostridium and Bacteroides) primers were employed in the analyses. The DGGE band pattern was subjected to cluster analysis to demonstrate the similarity and difference between dietary treatments and solid-liquid fractions. Fibre digestibility, gas production, and volatile fatty acid (VFA) concentration were increased with incubation time. The differences between solid and liquid fractions were great in total bacteria, Clostridium, and Bacteroides communities. Clostridium and Bacteroides communities appeared unaffected by incubation time, whereas great differences existed between solid and liquid fractions throughout incubations (experiment 1). Barley and kidney bean supplements increased gas production and lowered rumen fluid pH, whereas changes in VFA concentration were significant only for kidney bean supplement. The Clostridium, and Bacteroides groups were affected by barley and kidney beans in the liquid fraction. However, the solid fraction was stable (experiment 2). These results indicate a rigid and stable community structure of Clostridium and Bacteroides groups involved in the digestion of rice straw-based diets in the rumen. Distinctive differences between solid and liquid fractions were described; hence, separate analyses of two fractions may greatly help understand the relationship between fermentation patterns and microbial communities in the rumen. |

Sci., 2023, 68(8):356-363 | DOI: 10.17221/54/2023-CJAS The aim of this study was to monitor the occurrence of abnormal eggs in five initial laying lines of the domestic chicken (Gallus domesticus) between 2017 and 2021. The incidence of abnormal eggs, cracks, shell-less eggs, and double-yolk eggs were examined as part of the monitoring. Also assessed was the laying intensity, which ranged from 83.7% for the Barred Plymouth Rock (BPR) B line to 96.8% for the Rhode Island Red (RIR) A line. A statistically significant relationship between the line and control year was discovered for all varieties of abnormal eggs. The highest occurrence of cracks was in the RIR B line (2.2%). The BPR A line of hens had the highest concentration of shell-less eggs (1.7%), while the RIR A line, which also had the highest laying intensity (96.8%), had the highest frequency of double-yolk eggs (1.5%). A positive trend between the intensity and the occurrence of abnormal eggs was also confirmed. The BPR B (3.4%) and BPR C (3.0%) lines, which belonged to the lines with the lowest laying intensity, had the lowest percentage of abnormal eggs. For the BPR C line, cracks (1.2%), shell-less eggs (0.9%), and double-yolk eggs (0.9%) had the lowest occurrence rates. The correlation between laying intensity and double-yolk eggs reached 0.67, and a significant positive relationship was seen. On the other hand, no association was found between the laying intensity and the prevalence of cracked or shell-less eggs. There was also a strong correlation between abnormal eggs and double-yolk eggs (0.80) and between abnormal eggs and cracks (0.73). The correlation between the total occurrence of abnormal eggs and the intensity of laying reaches a mean value of 0.41. An increase in the intensity of laying from 80% to almost 100% leads to an increase in the occurrence of abnormal eggs from 2.6% to 6.3%. |

The results show that cows with CI1 shorter than 12 months have on average 2 345 kg lower milk yield in the first 1 000 days of productive lifetime than those with CI1 between 14 and 16 months. This is caused by lower MY in both the first and the second lactation as well as higher probability of being culled before reaching 1 000 days of productive life. |

Data set consisted of 770 865 records of gestation length in 375 574 Holstein cows and covered the period from 2012 to 2018. Mean gestation length was 277 ± 4.9 days, and it was 1.4 days longer in male calves compared to females, and 1.1 days longer in cows compared to heifers. Animal repeatability model with maternal effect was employed for variance component estimation. The direct genetic effect explained the highest proportion of variability, and it corresponded with moderate direct heritability (0.48), while maternal heritability was much lower (0.06). We estimated conventional and genomic breeding values with the genomic matrix based on 39 145 single nucleotide polymorphisms in 13 844 animals. Genomic breeding values were weakly (< 0.25) but significantly correlated with breeding values for type, production and fitness traits. Pearson correlations between breeding values indicated a negative association of direct gestation length with milk production, longevity and fertility of bulls, and a positive association of maternal gestation length with most of the type traits related to the body composition. Genetic trends for male and female parts of the population showed a tendency to the shortening of gestation, which should be of concern, as short gestation could be reflected in a negative indirect response in other correlated traits, such as the incidence of stillbirth, the health status of cows after calving, culling, or conception rate. Sci., 2020, 65(10):389-401 | DOI: 10.17221/165/2020-CJAS This study aimed to determine the associative effects (AEs) of 28 feed combinations of concentrate/soybean pod/alfalfa at different concentrate-roughage ratios that were incubated for 72 h in single tubes (120 ml) which were added 30 ml rumen buffered fluid. The gas production (GP) at 0, 2, 4, 6, 9, 12, 24, 36, 48, 72 h was recorded. A single exponential equation was applied to calculate the GP parameters a (rapid GP), b (slow GP), a + b (GP potential) and c (rate constant of slow GP that can reflect the specific process of GP, rapid and slow GP and GP rate). The AEs were calculated by 72 h GP and weighted estimation value of each combination. After 72 h incubation, pH, volatile fatty acids (VFA) and ammonia nitrogen (NH3-N), dry matter digestibility (DMD), organic matter digestibility (OMD) were determined the incubation fluid and residues. The single-factor AE index (SFAEI) and multiple-factor AE index (MFAEI) were computed. The results showed that the groups 50 : 50 : 0, 40 : 60 : 0, 60 : 20 : 20, 60 : 10 : 30, 50 : 30 : 20, 50 : 20 : 30, 40 : 50 : 10, 30 : 55 : 15, 30 : 40 : 30, 20 : 65 : 15, 20 : 50 : 30 had higher GP72 h, a, b, DMD, OMD, NH3-N, in addition, higher AE of GP, DMD, OMD, total VFA and NH3-N than those of the other groups (P < 0.05 or P < 0.01), especially the group 30 : 55 : 15 was optimal. In conclusion, in vitro data reveal reliable fermentability and the highest SFAEI and MFAEI occurred when concentrate, soybean pod and alfalfa were combined at the ratios of 50 : 50 : 0, 40 : 60 : 0, 60 : 20 : 20, 60 : 10 : 30, 50 : 30 : 20, 50 : 20 : 30, 40 : 50 : 10, 30 : 55 : 15, 30 : 40 : 30, 20 : 65 : 15, 20 : 50 : 30. Sci., 2020, 65(8):295-302 | DOI: 10.17221/14/2020-CJAS This study aimed to evaluate the effects of rectal temperature (RT) as well as artificial insemination (AI) technique on pregnancy rates in Holstein cows and artificial insemination or embryo transfer (ET) techniques in Holstein heifers during summer. The experiments were conducted on five dairy farms in Hungary, where 1 631 data were examined. The RT was measured immediately before reproductive techniques using a digital thermometer. The effects of animal group and month on RT were analysed. The enhancement of RT until September can be seen in heifers, but the highest RT of cows occurred during July. In line with the enhancement of RT the pregnancy rate decreased in all groups; the lowest (cows) and the highest (heifers) value was observed in July. Moderate to high negative coefficients of correlation were detected between rectal temperature and pregnancy rate for particular groups (r = -0.3 - -0.7). More than 92% of ET, 48% and 16% of inseminated heifers and cows were pregnant up to the category of 39.1 °C of RT, above this it did not change considerably in the inseminated groups. In inseminated animals with RT higher than 39.1 °C significantly higher services per conception could be observed contrary to ET heifers. ET may become a more effective strategy to improve pregnancy success in heifers compared to AI during summer. Czech J. Anim. Sci., 2020, 65(6):205-212 | DOI: 10.17221/108/2020-CJAS The study estimated phenotypic correlations between reproductive traits in sows classified as maternal (Polish Large White, Polish Landrace, and Yorkshire) and paternal (Duroc, Berkshire, and Hampshire) components, kept on farms located in Poland and in the United States. Altogether, it used data on 736 litters from 196 sows to analyse traits related to litter and reproductive cycle length. The former included litter size, the numbers and the percentages of piglets born alive, stillborn, and weaned; the latter included gestation length, lactation length, and the lengths of weaning-to-conception, farrowing-to-conception, and farrowing intervals. The strongest positive correlations between the litter-related traits were those between litter size and the number of piglets born alive (0.90), the number and the percentage of piglets born alive (0.88), the numbers of piglets born alive and weaned (0.78), and litter size and the number of weaned piglets (0.68); the strongest negative correlations were between the percentage of piglets born alive and the percentage (-0.95) and the number (-0.82) of stillborn piglets. Among the traits related to reproductive cycle length, the strongest positive correlations were those between the length of weaning-to-conception interval and the lengths of farrowing-to-conception (0.96) and farrowing (0.97) intervals, and between the length of farrowing-to-conception interval and farrowing interval (0.98). Gestation length and lactation length were weakly negatively correlated (-0.25). Correlation coefficients between the traits related to litter size from one side and reproductive cycle length from the other were low and mostly statistically non-significant, suggesting that using phenotypic correlations can help improve economically important reproductive traits, but only within the two above-mentioned groups of traits. This increases the likelihood of widespread genetic defects in livestock populations. Genome-wide studies have identified DNA loci associated with the disruption of foetal embryonic development and its death, which have been called "fertility haplotypes". The aim of this study is to analyse the occurrence of АН1 haplotype or rs475678587 in Ayrshire bulls (n = 186) used in the artificial insemination system of Russia and to evaluate the reproductive and productive qualities of their daughters. The proportion of genetic defect carrier bulls in the analysed sample cohort was 16.66%. Analysis of the proportion of carriers of unwanted haplotype depending on the place of birth of the bull for service showed that the highest frequency was observed in bulls of Canadian origin (26.66%). Based on data on 97 478 cow and heifer calvings, stillbirth and abortion rates were analysed and cow reproduction rates were based on an AH1 status for the bull-father and bull for service. We observed a significant increase (P < 0.05 and P < 0.001) in the incidence of stillbirths when crossing the daughters of AH1-C bulls with AH1-C bulls. There was no negative association of abortion rates and reproductive qualities with AH1 haplotype. The high incidence of AH1 haplotype among animals of Ayrshire breed requires regular screening for the carriers of this haplotype not only among bulls, but also in cows. Czech J. Anim. Sci., 2020, 65(6):213-223 | DOI: 10.17221/60/2020-CJAS Hermetia illucens (black soldier fly, BSF) larvae meal has shown to be a good protein source in monogastric animal diets, but published data regarding its immunomodulatory properties is limited. For this purpose, a study has been conducted to evaluate the effects of larvae meal on selected immune parameters and caecal bacterial counts of broiler quails. Hermetia illucens larvae were reared on two substrates (100% chicken feed, BSF-M, or 50% chicken feed + 50% fish offal, BSF-F) in order to manipulate the fatty acid profile of larvae meal by increasing the long-chain omega-3 (n-3) polyunsaturated fatty acids which are known for their immunomodulatory properties. For immunological purposes, a total of 60 birds were randomly allocated to three dietary treatment groups (n = 20/treatment): control quail diet (CON), or quail diets including 10% of BSF-F or BSF-M larvae meal. Blood was collected 27 and 37 days after the trial commenced to determine the humoral immune response, serum lysozyme concentrations, serum bactericidal activity and protein fractions of the serum. The dietary inclusion of BSF-M meal resulted in an increased secondary humoral immune response compared to the CON treatment group (P < 0.01). Quails in the BSF-F treatment had significantly higher serum lysozyme activity compared to quails in the CON and BSF-M treatment group. Both larvae meals significantly increased cell-mediated immunity on day 37 (P < 0.001) but they had no effect on serum bactericidal activity. Both larvae meal sources resulted in higher α2-globulin levels on both sampling days, whereas γ-globulin levels were significantly lower in quails with the BSF-F treatment on day 27. Dietary treatments had no significant effect on caecal bacterial counts. In conclusion, the present study showed that the larvae meal had immunostimulatory effects in quails, but the rearing substrate of larvae strongly influenced the immunostimulatory properties of the larvae meal. |
